摘  要:目的:检测帕金森病(PD)大鼠中缝背核与抑郁相关神经内分泌激素5-羟色胺(5-HT)、神经肽Y(NPY)、P物质(SP)蛋白及其mRNA的表达变化,探讨PD与抑郁的关系。方法:雄性SD大鼠单侧微量注射6-OHDA制备PD大鼠模型,将制模成功的10只雄性大鼠作为模型组,另10只正常大鼠作为对照组,采用免疫组织化学显色观察正常大鼠与PD大鼠中缝背核神经内分泌激素5-HT、NPY、SP阳性细胞的分布、形态及数量变化,采用RT-PCR方法在mRNA水平检测正常大鼠与PD大鼠中缝背核5-HT合成的限速酶-色氨酸羟化酶2(TPH2)、NPY和SP的表达变化。结果:免疫组织化学结果显示,模型组大鼠中缝背核5-HT、NPY免疫反应阳性神经元数量明显少于对照组,而模型组大鼠中缝背核SP免疫反应阳性神经元数量明显多于对照组。RT-PCR检测显示模型组大鼠中缝背核TPH2mRNA和NPYmRNA表达明显低于对照组,而模型组大鼠中缝背核SPmRNA表达明显高于对照组。结论:PD大鼠中缝背核与抑郁相关神经内分泌激素发生明显表达变化。Objective: To investigate the expression changes of depression-related neuroendocrine hormones serotonin (5-HT), neuropeptide Y (NPY), substance P (SP) and its mRNA in dorsal raphe nuclei of 6-OHDA induced Parkinson's disease rats, and to explore the relationship between PD and depression. Methods: Male adult Sprague-Dawley rats were microinjected with 6-OHDA into the right substantia nigra and medial forebrain bundle of the brain. After 2 weeks, there were 10 male model rats served as the model group. Six adult normal rats were injected saline instead of 6-OHDA as the control group. The expression of 5-HT, NPY and SP in dorsal raphe nucleus of rats in two groups were examined by immunohistochemical methods. The expressions of tryptophan hydroxylase 2 (TPH2 is 5-HT synthesis rate-limiting enzyme), NPY and SP at the rnRNA level were detected by RT-PCR. Results : The numbers of 5-HT and NPY immunoreactive cells in dorsal raphe nucleus of the model group were reduced significantly compared with those of the control group. The number of SP immunoreactive cells in dorsal raphe nucleus of the model group was increased significantly compared with those in the control group. The expressions of TPH2 mRNA and NPY mRNA in dorsal raphe nucleus of the model group were reduced significantly compared with those of the control group, while the expression of SP in dorsal raphe nuclei of the model group was increased significantly compared with that of the control group. Conclusion: The expression of depression-related neuroendocrine hormones significantly changed in dorsal raphe nucleus of 6-OHDA model rats with Parkinson's disease.
