摘  要:目的通过研究糖尿病小鼠角膜创伤愈合以及创伤后血管、中性粒细胞和神经纤维的改变,了解糖尿病对小鼠角膜创伤愈合的影响。方法 80只小鼠随机分成对照组(40只)和模型组(40只),应用链脲佐菌素诱导建立糖尿病模型。对模型组和对照组小鼠血糖、体质量进行监测。制作小鼠角膜上皮创伤模型,观察糖尿病小鼠角膜上皮创伤愈合情况,并对创伤后角膜缘血管、中性粒细胞分布情况进行观察。采用免疫荧光分析神经纤维在这两组小鼠角膜分布的差异。结果模型组在注射链脲佐菌素后24h,血糖升高至(17.1±0.5)mmol·L-1,此时对照组为(6.7±0.2)mmol·L-1(P<0.05)。6 d时模型组体质量显著降低至(28.2±0.9)g,2 d、3 d、4 d、5 d、6 d时组间血糖、体质量差异均有统计学意义(均为P<0.05),符合糖尿病改变。模型组小鼠创伤愈合时间比对照组延后5 d。模型小鼠角膜缘部位的血管纤细、稀疏,创伤后2 d对照组中性粒细胞为(212.3±6.3)个/视野,模型组为(184.7±3.2)个/视野,差异有统计学意义(P<0.05)。对照组的中央、旁中央、周边角膜神经纤维密度分别为(309.2±27.2)μm3·mm-3、(234.9±11.4)μm3·mm-3、(176.8±14.3)μm3·mm-3,模型组分别为(263.8±13.2)μm3·mm-3、(175.1±18.3)μm3·mm-3、(96.3±12.7)μm3·mm-3,差异均有统计学意义(均为P<0.05)。结论糖尿病会导致小鼠角膜上皮创伤愈合延迟,以及小鼠角膜血管、中性粒细胞及神经纤维的改变。Objective To investigate the effects of diabetes on corneal wound healing through observing the changes of wound healing, distribution of vascular, neu- trophils and nerve fibers in mice. Hethods A total of 80 male mice were randomly di- vided into control group and diabetes group,40 cases in each group. The diabetic mod- els were established using streptozotocin (STZ) before the experiment. After that, the blood glucose and body weight of two groups were monitored, the corneal epithelial wound models were established to evaluate the corneal healing capability, the changes of limbal vascular and neutrophils were observed, and the distribution of corneal nerve fibers of each group were assessed by immunofluorescence. Results At 24 hours after STZ injection, the blood glucose increased to ( 17.1 ± 0.5 ) mmol ·L^-1 in diabetes group, which in control group was(6.7 ±0.2 )mmol·L^-1 ,there was statistical difference(P 〈 0.05 ). The body weight of diabetes group at 6 days was significantly reduced to( 28.2 ± 0.9) g,there were statistical differences in blood glucose and body weight at 2 days, 3 days,4 days,5 days,6 days between two groups ( all P 〈 0.05 ). Wound healing in diabe- tic mice in diabetes group was delayed for five days compared with the control group. The limbal vascular of diabetic mice was slender and sparse. Neutrophils in control group were( 212.3 ± 6.3 )cells per visual field at the 2 days ,which in the diabetes group was ( 184.7 ± 3.2 ) cells per visual field, there was statistical difference ( P 〈 0. 05 ). In control group, the nerve fibers densities in central, paracentral, peripheral of cornea were (309.2±27.2)μm3·mm^-3,(234.9± 11.4)μm3·mm^-3,(176.8±14.3)μm3·mm^-3 ,respectively, which in diabetes group were (253. 8 ± 13. 23 )μm3·mm^-3, (175.1 ± 18.3)μm3·mm^-3, (96.3 ± 12.7)μm3·mm^-3 ,respectively,there were statis- tical differences ( all P 〈 0.05 ).
