摘　　要：目的探讨丙戊酸钠联合As2O3对NB4细胞系血管内皮生长因子(VEGF)、核因子-κB(NF-κB)和基质金属蛋白酶9(MMP-9)表达的影响。方法实验分对照组、As2O3组、VPA组和As2O3联合VPA组,四甲基偶氮唑蓝(MTT)法检测细胞存活率,以RT-PCR方法检测VEGF、NF-κB和MMP-9 mRNA的表达,以Western-blot方法检测VEGF、NF-κB和MMP-9蛋白的表达。结果 VPA增加As2O3诱导NB4细胞系存活率下降,NB4细胞在As2O3作用下VEGF、NF-κB和MMP-9表达增高(P<0.05)。联合VPA组3者表达均比单用As2O3组降低(P<0.05)。结论VPA可下调As2O3引起的VEGF、、NF-κB和MMP-9表达增高。Objective To investigate the expressions of vascular endothelial growth factor, NF-KB and MMP-9 in NB4 cell lines affected by VPA in combination with arsenic trioxide. Methods The experiment included control group, arsenic trioxide group, VPA group and group of arsenic trioxide plus VPA, MTF assay method was used to detect the cell survival rate. RT-PCR was used to detect the mRNA level of VEGF, NF-KB and MMP-9. Western Blot was used to detect the protein level of VEGF,NF-KB and MMP-9. Results VPA facilited the survival de creased rate induced by arsenic trioxide in NB4 cell lines, the expression of VEGF, NF-KB and MMP-9 in NB4 cell lines were increased under the effect of arsenic trioxide （ P 〈 0.05 ）. The expression of VEGF, NF-KB and MMP-9 in the group of VPA plus arsenic trioxide were all decreased than in the group of arsenic trioxide alone （P 〈 0. 05 ）. Conclusions VPA down-regulates the increased expression of VEGF, NF-KB and MMP-9 caused by arsenic trioxide.
