摘 要:目的:调查贵州苗族、布依族、侗族人群mtDNA的群体遗传特点。方法:用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术和DNA测序法,对3个民族男性个体mtDNA上12个SNP进行多态性分析。结果:285名个体共检出47种单倍型和12种单倍群。主要单倍型H32、H36、H10在3个民族间频率分布差异有统计学意义(P<0.05);3个民族共有单倍型为11种(72.98%)且在民族间频率分布差异有统计学意义(P<0.05),单倍群B(21.05%)和M7(21.05%)频率最高,B在苗族、布依族中分布频率与侗族间差异有统计学意义(P<0.05)。结论:贵州苗族、布依族、侗族人群mtDNA单倍型分布相似,单倍群频率及主成分分析表现出南方群体特征。侗族母系遗传结构较苗族和布依族更为复杂,民族间存在基因交流的可能。Objective:To study the population genetics characteristics of mitochondrial DNA in ethnic Miao, Buyi and Dong populations from Guihzou province. Method:Polymerase chain reaction and restriciton fragment length polymorphism (PCR - RFLP) was used to de- tect the 12 SNP loci in a total of 285 samples. The result was verified with DNA sequencing. Result:The 47 haplotypes and 12 haplogroups were detected in 285 samples. The main haplotypes were H32, H36 and H10 (41.04%), but the difference was statistically significant a- mong the three ethnic groups (P 〈 0.05). There were 11 haplotypes were found in all the groups. By the way, H11 was only found in Miao population (11.58% ). Haplogroup B and M7 were the predominant types (both of 21.05% ). In haplogroup B, Miao and Buyi had differ- ence with Dong respectively and the difference was statistically significant (P 〈 0. 05 ). Conclusion: There was greatly similarity in the dis- tribution of the haplotype of the mtDNA among the three ethnic groups, but there were some difference in the distribution of haplo- types. Haplogroups and Principal Component (PC) analysis showed that the genetic polymorphism of mtDNA of the three populations indi- cated their origination from southern China. The matrilineal genetic structure of Dong population is more complex than the others, but they all have the possibility of gene flow.
