摘 要:目的:探讨肌球蛋白轻链激酶(myosin light chain kinase,MYLK)和肌球蛋白轻链(myosin regulatory light chain9,MYL9)在非小细胞肺癌(non-small cell lung cancer,NSCLC)组织的表达情况及临床意义。方法:采用qRT-PCR及蛋白质印迹法检测2012-08-20-2013-02-01广西医科大学第一附属医院胸外科30例NSCLC组织和对应的癌旁组织及30例正常肺组织中MYLK和MYL9mRNA及蛋白的表达情况。进一步对影响MYLK和MYL9表达的临床特征进行Logistic回归分析。结果:NSCLC癌组织MYLK基因相对表达量为0.50±0.19,癌旁组织为0.97±0.22,正常组织为1,癌组织低于癌旁(P=0.001)及正常组织(P<0.001);MYLK蛋白在肺癌组织、癌旁组织及正常组织中表达分别为0.54±0.18、0.72±0.21和0.84±0.23,癌组织低于癌旁(P=0.008)及正常组织(P=0.003);MYL9基因在肺癌组织、癌旁组织及正常组织相对表达量分别为0.37±0.12、0.95±0.19和1,癌组织低于癌旁(P<0.001)及正常组织(P<0.001);MYL9蛋白在肺癌组织、癌旁组织及正常组织中蛋白相对表达量分别为0.56±0.14、0.74±0.20和0.78±0.19,癌组织低于癌旁(P=0.001)及正常组织(P<0.001)。MYLK和MYL9的mRNA及蛋白相对表达量与病理类型、分化程度和临床分期无关。癌组织淋巴结转移组MYLK的mRNA(P=0.04)和蛋白(P=0.04)及MYL9的mRNA(P<0.001)和蛋白(P=0.024)相对表达量明显高于无淋巴结转移组;吸烟组MYLK的mRNA(P<0.001)和蛋白(P=0.038)及MYL9的mRNA(P=0.001)及蛋白(P=0.01)相对表达量低于无吸烟组;男性MYLK的mRNA(P<0.001)和蛋白(P=0.03)及MYL9的mRNA(P=0.001)和蛋白(P<0.001)相对表达量低于女性组。多因素分析显示,女性、无吸烟及淋巴结转移增加MYLK及MYL9的表达。结论:NSCLC中MYLK和MYL9mRNA及蛋白表达均明显低于癌旁及正常组织,且在淋巴结转移的癌组织中表达均高于无淋巴结转移组。女性、无吸烟及淋巴结转移是MYLK及MYL9表达量增高的主要影响因素。OBJECTIVE: To explore the clinical significance of MYLK and MYL9 expression in non- small cell lung cancer(NSCLC). METHODS: Thirty cases of non-small cell lung cancer tissue, corresponding paraneoplastic tissues and 30 cases of normal lung tissues from 2012-08-20 to 2013-02-01 in department of cardiothoracic surgery of First Affiliated Hospital of Guangxi Medical University were detected by quantitative Real-time PCR and Western blot methods. The clin- ical features of expression of MYLK and MYL9 were analyzed by Logistic regression analysis. RESCLTS.. The relative ex- pression of MYLK mRNA and protein in non-small cell lung cancer was 0.50±0.19 and 0. 54±0.18,which was signifi- cantly lower than that in the paraneoplastic tissues (0.97 ± 0.22,0. 72 ± 0.21) and normal tissues ( 1,0. 84 ± 0.23, all P values 〈0.05),paraneoplastic tissues and normal tissues were found no statistically significant difference(P〉0.05). While in the MYL9,the relative expression of MYL9 mRNA and protein in non-small cell lung cancer was 0. 37±0.12 and 0.56±0.14 ,which was significantly lower than that in the paraneoplastic tissues (0. 95±0.19,0. 74±0.20) and nor- mal tissues (1,0.78±0.19, all P〈0.05), paraneoplastic tissues and normal tissues were found no statistically significant difference(all P〉0.05). MYLK mRNA and MYL9 mRNA in lymph node metastasis and non- metastasis (MYLK:P= 0.04;MYL9:P〈0.001),smoking and non-smoking(MYLK:P〈0. 001; MYL9: P= 0. 001) and male and female (MYLK:P〈0. 001 ;MYL9:P=0. 001) were statistically significant different,whereas in MYLK and MYL9 protein ex- pression in lymph node metastasis and non- metastasis( MYLK: P= 0.04 ; MYL9 : P = 0. 024 ), smoking and non-smoking (MYLK:P=0. 038;MYL9:P=0.01) and male and female (MYLK:P=0.03;MYL9:P〈0. 001). The multiple regres- sion analysis showed that only female, non-smoking and lymphatic node metastasis increased MYLK and MYL9 expression markedly. CONCLUSIONS: Com
