摘  要:目的观察颈内动脉移植骨髓间充质干细胞(bonemarrowstemcells,BMSCs)透过血脑屏障在血管性痴呆(vasculardementia,VD)大鼠脑内存活、迁移及其对海马细胞凋亡和认知能力的影响。方法取幼年雄性清洁Wistar大鼠股骨骨髓单个核细胞(BMNCs),贴壁培养后,取3—4代BMSCs诱导分化并BrdU标记。制作VD大鼠模型,术后24h颈内动脉移植0.5m11.2×10V/ml的BrdU标记细胞。术后2、4、8周检测移植细胞在VD大鼠脑内存活、迁移以及大鼠学习记忆能力,并检测大鼠海马细胞凋亡情况。结果颈内动脉移植后BMSCs广泛存活于VD大鼠脑组织内,随着时间的延长,迁移方向集中,主要聚集至海马、大脑皮质等缺血易损伤区域;术后2、4、8周模型组大鼠AAR比率低于对照组,差异非常显著(P〈0.01);治疗组较模型组比例明显提高(P〈0.01)。治疗组大鼠术后各时相点海马细胞凋亡比例显著低于模型组(P〈0.01)。结论BMSCs颈内动脉移植后可透过血脑屏障向海马、大脑皮质等脑缺血易损伤区迁移聚集、存活,显著提高VD大鼠的认知能力,并对大鼠海马区神经细胞具有明显保护作用。To investigate survival and migration intracarotid-transplanted bone marrow stem cells (BMSCs) in vascular dementia (VD) rats and its impact on hippoeampus apoptosis and cognitive abilities, BMSCs were isolated from bone marrow in vitro, labeled with BrdU and injected into vascular dementia rats through intracarotid at 1.2×107 cells/ml of 0.5 ml. 2, 4 and 8 weeks later after the transplantation, BMSCs were observed throughout the brain tissue and the vast majority distributed in the ischemic zone active avoidance response (AAR) ratio in the model group of injury such as hippocampus and cerebral cortex. The showed a significant decrease compared to the control group (P〈0.01). BMSCs treatment significantly increased the AAR ratio. Collectively, Intracarotid transplantation of bone marrow stem cells could enter the VD rats cerebra parenchyma across blood-brain barrier (BBB), migrate into the damaged brain tissue to localize, survive, and significantly improved the cognitive abilities of vascular dementia rats, which indicates that BMSCs therapy has certain protection on cerebral histiocytes.
