肝癌患者血浆中PLAG1的异常表达

摘　　要：目的:本研究旨在探讨多形性腺瘤基因1(pleomorphic adenoma gene 1,PLAG1)在肝细胞癌患者血浆中的异常表达及其临床意义.方法:收集42例肝癌患者、34例肝硬化患者及30例健康人血浆,应用SYBR GreenⅠ实时荧光定量PCR技术检测PLAG1 mRNA在健康人、肝硬化患者及肝癌患者血浆中的表达情况,并分析其与肝癌患者临床病理特征之间的关系.结果:SYBR GreenⅠ实时荧光定量PCR结果显示,PLAG1 mRNA在肝癌组、肝硬化组及健康体检组的血浆中阳性表达率分别为:71.4%、23.5%和6.7%.PLAG1 mRNA在肝癌组血浆中的阳性表达率明显高于健康体检组及肝硬化组(P<0.05).而健康体检组和肝硬化组血浆中PLAG1 mRNA阳性表达率差异无统计学意义(P>0.05).同时对肝癌组、肝硬化组及健康体检组中PLAG1 mRNA的扩增产物进行相对定量的分析比对,发现三组间比较差异有统计学意义(P<0.05).肝癌患者血浆中PLAG1 mRNA表达水平与肝癌的病理分化程度及远处转移有显著的关系(P<0.05).结论:PLAG1 mRNA在肝癌患者血浆中的表达,提示其可能在肝癌的发生发展过程中发挥重要的作用,有望成为新的辅助诊断肝癌,判断转移风险的临床检测指标.AIM： To investigate the abnormal expression of pleomorphic adenoma gene 1 （PLAG1） in plasma of patients with hepatocellular carcinoma （HCC） and to analyze its clinical significance. METHODS： Plasma samples were collected from 42 HCC patients, 34 liver cirrhosis patients and 30 healthy donors. The mRNA expression levels of PLAG1 were quantified by SYBR Green I real- time fluorescent quantitative PCR. The correlation between clinicopathologic characteristics of HCC and expression of PLAG1 was evaluated. RESULTS： The positive rate of PLAG1 mRNA in the HCC group, liver cirrhosis group, and healthy control group were 71.4%, 23.5% and 6.7%, respectively. PLAG1 mRNA expression level in the HCC group was significantly higher than those in other two groups （P 〈 0.05 for both）. However, no significant difference was found between the liver cirrhosis group and healthy control group （P 〉 0.05）. Significant difference in quantitative data from SYBR Green I real-time fluorescent quantitative PCR was also found among the three groups （P 〈 0.05）. PLAG1 mRNA expression level in HCC patients was significantly correlated with pathological grade and tumor metastasis （P 〈 0.05 for both）. CONCLUSION： The expression level of PLAG1 mRNA in plasma of HCC patients may play a significant role in the pathogenesis and development of HCC, and it may serve as a novel tumor marker for diagnosis and metastasis of HCC.
