摘 要:目的研究细胞松弛素B(CB)对人胶质瘤细胞系U87细胞增殖及凋亡的影响及机制。方法采用CCK-8试剂盒检测不同浓度的CB对胶质瘤细胞系U87细胞增殖的影响并得出一个最适时间及半抑制浓度(IC50)。观察最适半抑制浓度处理的细胞其透射电镜下的细胞形态及微丝改变。流式细胞术Annexin-Ⅴ-FITC/PI双染检测U87细胞的凋亡情况。结果不同浓度的CB对U87细胞的抑制效应呈时间及剂量依赖性。CB作用于U87细胞的最适作用时间为72 h,其最适半抑制浓度为2.694×10-5mol/mL。可诱导(12.667±1.159)%的细胞凋亡。电镜显示细胞内微丝结构,微丝断裂,出现大量短棒状微丝结构。结论 CB可能是通过破坏胶质瘤细胞质内微丝结构抑制U87细胞增殖,促进胶质瘤细胞凋亡。Objective To investigate the role of cytochalasin B (CB) on proliferation and ap- optosis of human glioma cell line U87 cells and its mechanism. Methods CCK-8 method was used to detect different concentrations of CB on the proliferation of U87 glioma cells and to ob- tain an optimal time and half inhibitory concentration ( ICs0 ). The optimal half inhibitory con- centration on cells morphology and the microfilament changes by transmission electron micro- scope was observed. Apoptosis of U87 cells were detected by flow cytometry Annexin-V-FITC/ PI double staining. Results Inhibitory effect of different concentration of CB on U87 cells was dose and time dependent. The optimum effects time of CB on U87 cell was 72 h, and the half inhibitory concentration was 2. 694 x 10-5 mol/mL. It induced ( 12. 667 ± 1. 159)% cells ap- optosis. Electron microscopy showed intracellular microfilament fracture and appeared a lot of short rod-like microfilaments. Conclusion CB can inhibit the U87 cells proliferation, which may through the destruction of glioma cells' microfilament, so as to promote the increasing of glioma cell apoptosis rate.
