基质金属蛋白酶-9基因在食管鳞癌患者中的表达

摘　　要：目的分析基质金属蛋白酶-9(MMP-9)基因在食管鳞癌患者中的表达及二者的关系。方法选自2010年5月—2012年9月在河北省卢龙县医院治疗的90例食管鳞癌患者。同时选取健康检查者50例作为正常对照组,对比两组的MMP-9基因表达水平,并分析临床各指标与MMP-9基因表达的关系。结果 MMP-9在正常食管黏膜表达的阳性率为8.0%(4/50),而在食管鳞癌组织表达的阳性率为62.2%(56/90),两者的MMP-9表达阳性率差异有统计学意义(P<0.05);MMP-9表达与年龄、性别、肿瘤部位、组织学类型、组织分化程度差异均无统计学意义(P>0.05),MMP-9表达与TNM分期、是否存在淋巴结转移具有一定的关系(P<0.05);MMP-9表达与MMP-2表达差异有统计学意义(χ2=5.315,P<0.05),而MMP-9表达与细胞间黏附分子-1(ICAM-1)、转录因子p50(NF-κB p50)、环氨酶-2(COX-2)表达差异无统计学意义(P>0.05)。结论 MMP-9在食管鳞癌患者的表达高于正常食管黏膜中的表达,而且MMP-9表达与食管鳞癌患者发病年龄、性别、T分期、组织分化程度无明显关系,但与TNM分期、是否存在淋巴结转移具有一定的关系,与ICAM-1、NF-κB p50、COX-2也存在着一定的关系。[ Objective] to analyze the correlation of expression of matrix metalloproteinase-9 （MMP-9 ） in patients with esophageal squamous carcinoma and their relationship. [ Methods ] A total of 90 cases of esophageal squamous cell carcinomas treated in Lu long People＇s Hospital of Hebei Province from May 2010 to September 2010 were selected. Another 50 cases of healthy physical examination were selected as normal control group, then gene expression levels of MMP-9 were compared between 2 groups, and the correlation between MMP-9 gene and others clinical factors was analyzed. [ Results] The positive expression rate of MMP-9 in nor- mal esophageal mucosa and esophageal squamous carcinoma tissue was 8.0% （4/50 } and 62.2% （56/90）, respectively, with sig-nificant difference （ P 〈 0.05 ）. Expression of MMP-9 was not correlated with age, gender, tumor location, histological type, differ entiation degree （ P 〉 0.05 ）, while it had a certain relationship with TNM stage and the existence of lymph node metastasis （ P 〈 O. 05 ）. It is significantly different between MMP-9 expression and the expression of MMP-2 （ x2 = 5.315, P 〈 0. 05 ） , and the expression of MMP-9 was not correlated with ICAM-1, the NF-KB pS0 and cox-2 expression （ P 〉 0.05 ）. [ Conclusion] The expres-ion level of MMP-9 in esophageal squamous carcinoma is higher than its expression level in normal esophageal mucosa, and the ex-pression of MMP-9 is not obviously correlated with esophageal squamous carcinoma onset age, sex, T stage, tissue differentiation degree, but is related with TNM staging, the existence of lymph node metastasis, as well as MMP - 9 and ICAM-1 ,NF-KB p50, COX-2.
