摘 要:目的:观察黄连解毒汤对慢性束缚应激刺激联合D-半乳糖致拟阿尔茨海默病小鼠行为学和脑组织Ach、TchE以及血清NE、5-HT等神经递质的影响。方法:BALB/c小鼠56只,随机分为4组:正常对照组、D-半乳糖组、复合模型组、黄连解毒汤组。复合模型组及黄连解毒汤组均给予慢性束缚应激刺激,黄连解毒汤组同时予黄连解毒汤6 g/kg灌胃干预,连续14 d。实验期内D-半乳糖组、复合模型组和黄连解毒汤组均采用D-半乳糖120 mg/kg颈背部皮下注射,1次/d,连续28 d。实验结束后,进行旷场试验,并检测各组小鼠脑组织Ach、TchE和血清NE、5-HT的含量。结果:与正常对照组和D-半乳糖组比较,复合模型组小鼠中央区域路程和总路程均减少,脑组织Ach和血清5-HT、NE明显降低,脑组织TchE明显增加;与复合模型组比较,黄连解毒汤组小鼠中央区域路程和总路程均增加,脑组织Ach、血清5-HT、NE升高,脑组织TchE减少。结论:黄连解毒汤能明显改善慢性束缚应激刺激联合D-半乳糖致拟阿尔茨海默病小鼠行为学异常,升高小鼠脑组织Ach和血清NE、5-HT等神经递质含量。Objective: To observe the effects of Coptis detoxication decoction( CDD) on behaviors,Ach,TchE in brain tissue,serum levels of NE,5-HT in chronic mild stress and D-galactose induced Alzheimer disease like mouse model. Methods: Fifty-six BALB / c Mice were divided randomly into four groups: control group,D-galactose group,compound model group and CDD group. Compound model group and CDD group were subjected to chronic stress and CDD group was intragastrically infused with 6 g / kg CDD for 14 days. At the same time,mouse in D-galactose group,compound model group and CDD group were injected 120 mg / kg D-galactose once a day from the 1st day to 28th day. Then open-field test was experienced and contents of Ach and TchE in brain tissue,serum levels of NE and 5-HT were detected in all groups. Results: Compared with those of control and D-galactose group,the central area distance and total distance of compound model group were shortened,Ach in brain tissue,serum levels of 5-HT and NE were decreased markedly,while TchE in brain tissue was significantly increased. Compared with those of compound model group,the central area distance and total distance of CDD group were prolonged,Ach in brain tissue,serum levels of 5-HT and NE were increased,while TchE in brain tissue was decreased. Conclusions: CDD can significantly improve the behavior of chronic mild stress and D-galactose induced Alzheimer disease like mouse model,increasing neurotransmitters of Ach in brain tissue and serum levels of NE and 5-HT.
