HPV和CDK6与宫颈癌的相关性研究

摘　　要：目的研究人乳头瘤病毒E7（HPVE7）蛋白和细胞周期蛋白依赖激酶6（CDK6）蛋白在宫颈癌组织中的表达及其与临床病理各因素之间的关系。方法采用免疫组化（S-P）方法检测30例宫颈癌、30例宫颈上皮内瘤变及30例正常宫颈组织中HPVE7蛋白和CDK6蛋白的表达情况。结果HPVE7蛋白表达阳性率在正常宫颈组织中低于CIN组，差别有统计学意义（X^2=4．55，P〈0．05），在CIN组低于宫颈癌组，差别有统计学意义（X^2=4．28，P〈0．05）；CDK6蛋白表达阳性率在正常宫颈组织中低于C1N组，差别有统计学意义（X^2=5．97，P〈0．05），在CIN组低于宫颈癌组，差别有统计学意义（X^2=5．40，P〈0．05）；在宫颈癌组中CDK6蛋白表达与HPVE7蛋白表达存在正相关（r=0．563，P〈0．05）。结论HPVE7蛋白和CDK6蛋白在宫颈癌组织中表达升高，在宫颈癌的发生发展及侵袭中起重要作用，二者在宫颈癌的发生发展中起到协同作用。Objective To determine the expressions of human papillomavirus E7 （HPV E7） and cyclin-dependent kinases 6 （CDK6） in human cervical cancer tissue and explore their relationships with clinicopathology. Methods The expressions of HPV E7 and CDK6 in tissues of 30 cases of cervical cancer, 30 cases of CIN and 30 normal cases were detected by using immunohistochemical （ S - P） method. Results The expression level of HPV E7 in CIN cases was higher than that in normal cases （X^2 =4.55 ,P 〈0.05）, but it was lower than in cervical cancer cases （X^2 = 4.28,P 〈 0.05 ）. The expression level of CDK6 in CIN cases was higher than that in normal cases （X^2 = 5.97, P 〈 0.05 ） , but it was lower than in cervical cancer cases （X^2 = 5.40, P 〈 0.05 ）. In cervical cases the expression of CDK6 was positively correlated with that of HPV E7 （ r = 0. 563, P 〈 0.05 ）. Conclusion The expressions of HPV E7 and CDK6 in cervical cancer increase, which plays an important role in the carcinogenesis, development and invasion of cervical cancer. Furthermore, HPV E7 and CDK6 have synergetic effect on development of cervical cancer.
