摘  要:目的观察转化生长因子β1(TGF-β1)抗体对二氧化硅(SiO2)刺激的小鼠肺成纤维细胞(L929细胞)TGF-β1及Ⅰ型胶原表达的影响,探讨外源性TGF-β1抗体在抗肺纤维化中的作用。方法将L929细胞分为对照组、SiO2组和TGF-β1抗体组。对照组只加入RPMI-1640培养液3 ml,SiO2组、TGF-β1抗体组分别加入质量浓度为100.0mg/L的SiO23 ml,间隔2 h后,TGF-β1抗体组再加入质量浓度为1.0 mg/L的TGF-β1抗体1 ml,培养24 h后,采用酶联免疫吸附测定法检测L929细胞TGF-β1水平,免疫组化法检测TGF-β1及Ⅰ型胶原蛋白表达水平。结果 3组TGF-β1水平及蛋白表达水平分别比较,差异均有统计学意义(P<0.05);SiO2组TGF-β1水平和蛋白表达水平均高于对照组[(16.15±1.10)vs(9.15±0.44),(73.81±10.80)vs(18.42±3.48),P<0.05],TGF-β1抗体组TGF-β1水平和蛋白表达水平均低于SiO2组[(12.56±0.36)vs(16.15±1.10),(52.09±9.43)vs(73.81±10.80),P<0.05]。3组间Ⅰ型胶原表达水平比较,差异有统计学意义(P<0.05);SiO2组Ⅰ型胶原表达水平高于对照组[(42.36±8.34)vs(7.64±2.67),P<0.05],TGF-β1抗体组Ⅰ型胶原表达水平低于SiO2组[(29.81±7.45)vs(42.36±8.34),P<0.05]。结论 TGF-β1抗体可降低L929细胞中TGF-β1水平,并降低TGF-β1蛋白及Ⅰ型胶原蛋白的表达。Objective To observe the effects of transforming growth factor-β1 ( TGF-β1 ) antibody on the content of TGF-β1 and the expression of TGF-β1 and type Ⅰ collagen in mouse lung fibroblast cell (L929 cell) stimulated by silicon dio-xide (SiO2) , and explore its role of anti-fibrosis. Methods L929 cells were separated into three groups : control group, SiO2 group and TGF-β1 antibody group. RPMI-1640 medium (3 ml) was only added to the control group, SiO2 at the concentra-tion of 100. 0 mg/L (3 ml) were added to the SiO2 group and the TGF-β1 antibody group respectively. Two hours later, TGF-β1 antibody at the concentration of 1.0 mg/L (l ml) was added to the TGF-β1 antibody group. After 24 hours incu-bation, the level of TGF-β1 in L929 ceils was detected with the enzyme linked immunosorbent assay, and the protein expre- ssions of TGF-β1 and type Ⅰ collagen were examined by immunological method. Results There were significant diffe-rences in the levels and protein expressions of TGF-β1 among different groups (P 〈 0. 05 ) , which were evidently higher in SiO2 group than those in the control group [ ( 16. 15 ± 1.10) vs ( 9. 15 ± 0. 44), (73.81 ± 10. 80) vs ( 18.42 ± 3.48 ), P 〈0. 05], while which in TGF-β1 antibody group decreased significantly, when compared with SiO2 group [ (12. 56 ± 0. 36) vs ( 16. 15 ± 1.10), (52. 09 ± 9.43 ) vs (73.81 ± 10. 80), P 〈 0. 051. The protein expressions of type Ⅰ collagen among 3 groups were significantly different ( P 〈 0. 05 ), which in SiO: group increased significantly when compared with the control group[ (42. 36 ±8.34) vs (7.64 ± 2. 67), P 〈 0. 05], while in TGF-β1 antibody group it was significantly lower than that in SiO2 group [ (29. 81 ± 7.45) vs (42. 36 ± 8.34), P 〈 0. 05 ]. Conclusion TGF-β1 antibody could de-crease the TGF-β1 level and reduce the protein expressions of TGF-β1 and type I collagen in mouse lung fibroblast to alle-viate the lung fibro
