摘 要:海湾扇贝是雌雄同体动物,自体受精给杂交育种带来了极大的困扰。为提高杂交效率,本文实验用药物控制海湾扇贝自体受精并研究将其应用于育种实践的方法。首先用不同浓度的试剂A处理海湾扇贝的精和卵60min,结果表明随着试剂A浓度的增加,海湾扇贝受精率明显受到抑制,能够完全抑制受精的最低有效处理为用0.05%试剂A处理3分钟。处理后卵子的大部分受精能力可以通过反复洗卵恢复,0.1%的试剂B处理只对于用试剂A处理时间较长卵子的受精能力恢复有作用,但更高浓度则抑制受精能力。本文的实验证明,用0.05%试剂A的处理3分钟可完全抑制自体受精,且卵子的受精能力可通过洗卵后有效恢复,大部分受精卵可正常发育。因此,可以通过药物来控制海湾扇贝的自体受精来获得有正常受精能力的未受精卵。Self-fertilization caused tremendous problems in the cross breeding of hermaphroditic animals such as the bay scallop,Argopecten irradians.To increase the efficiency of cross breeding,we studied the prevention of self-fertilization using chemicals and its potential application in cross breeding.We first treated the mixed eggs and sperm with increasing concentrations of Reagent A and found that the self-fertilization rate decreased with the increase of Reagent A concentration.The minimum treatment of Reagent A to block self-fertilization is at 0.05 % for 3 min.The fertilization capability of eggs after such treatment can be largely recovered by washing with fresh seawater,but not by Reagent B,the antagonist of Reagent A.Most of the resulted fertilized eggs can develop normally.Therefore,we are able to prevent self-fertilization and obtain unfertilized eggs with normal fertilization capability with chemicals.The method developed here may be immediately applicable in cross breeding of hermaphroditic bivalves.
