摘 要:目的 研究单纯疱疹病毒 1型 (HSV 1)对L6 5 6 5小鼠白血病病毒 (L6 5 6 5MLV)诱发L6 5 6 5小鼠白血病的影响。方法 设L6 5 6 5MLV单独感染组、L6 5 6 5MLV和HSV 1共同感染组及正常对照组。两感染组小鼠分别用L6 5 6 5MLV、L6 5 6 5MLV和HSV 1(紫外线灭活 )经腹腔注射感染 ,观察小鼠感染后的发病情况 ,并用逆转录 聚合酶链反应 (RT PCR)检测L6 5 6 5MLVRNA在小鼠体内的分布。共连续观察 16周。结果 L6 5 6 5MLV单独感染组小鼠接种病毒后第 2周 ,首先在小鼠胸腺和脾脏检测到L6 5 6 5MLVRNA ,于第 8周后可在多种脏器中检测到病毒核酸。部分小鼠于接种病毒后10~ 12周出现早期白血病改变 ,发病率为 42 % ;L6 5 6 5MLV和HSV 1共同感染组小鼠接种病毒后第1周即在小鼠胸腺查到L6 5 6 5MLVRNA ,接种后第 2周病毒核酸出现在小鼠脾脏和外周血中 ,第 6周在多种脏器中可检测到病毒RNA。部分小鼠于接种病毒后 7~ 8周出现早期白血病改变 (有 1只小鼠发生白血病 ) ,发病率为 74%。正常对照组无小鼠发病。结论 HSV 1可能促发L6 5 6Objective Numerous studies have shown that herpes simplex virus type 1 and 2 (HSV 1 and HSV 2) are able to activate an endogenous type C virus of mouse. However, the influences of HSV 1 or HSV 2 on the disease induced by type C virus (such as retrovirus) are not clear. This experimental study was designed to understand the influence of HSV 1 on the L6565 mouse leukemia induced by L6565 MLV. Methods Ninety two suckling mice were randomly divided into 3 groups, i.e., group 1 inoculated with L6565 MLV alone (36 mice) group 2 inoculated with L6565 MLV plus HSV 1 group (35 mice), and group 3, control group without any inoculation (21 mice). After inoculations, two mice from each group were sacrificed every week. The peripheral blood, thymus, lymph node, spleen and liver were removed from each mouse. The tissue pathological changes were observed and the L6565 MLV RNA was detected by using RT PCR. Results In group 1, L6565 MLV RNA was detected firstly in mouse thymus and spleen at the second week after inoculation, and the viral RNA was detected in many murine organs at the 8th week post inoculation. The early phase of leukemia was induced in some mice 10 12 weeks post L6565 MLV inoculation, the incidence was 42%. In group 2, L6565 MLV RNA was detected in murine thymus at the first week after inoculation. Then, the viral RNA was detected in murine spleens and peripheral blood at the 2nd week post inoculation, and the viral RNA was detected in many murine organs at the 6th week post inoculation. The early phase of leukemia was induced in mice 7~8 weeks post L6565 MLV plus HSV 1 inoculation, the incidence was 74%. Conclusion HSV 1 could accelerate the L6565 mouse leukemia induced by L6565 MLV.
