凋亡调控基因在急性白血病的表达及其临床意义

摘　　要：研究凋亡调控基因 bcl- 2和 bax m RNA在急性白血病 ( AL)中的表达、相互关系和临床意义 ,应用半定量逆转录 -聚合酶链反应 ( RT- PCR)和 Southern blot技术检测 3 2例 AL患儿 bcl- 2和 bax m RNA水平及其比值。结果显示 ,AL患儿骨髓细胞中 bcl- 2 m RNA的表达明显高于对照组 ( P<0 .0 1) ,bax m RNA的表达明显降低( P<0 .0 1)。治疗后 ,bcl- 2 m RNA在两组间的表达无差异 ( P>0 .0 5 )。分析 bcl- 2 /baxm RNA表达比值显示 ,高比值组 NR率显著高于低比值组 ( P<0 .0 1)。结论 :AL的发病与凋亡调控基因 bcl- 2和 bax的异常表达有关。bcl- 2和 bax m RNA及其比值可作为判断To investigate the expression, correlation and clinical significance of bcl 2 and bax genes in AL, using the method of semi quantitative reverse transcription polymerase chain reaction (RT PCR) and Southern blot were used to the level and ratio of bcl 2 and bax mRNA in 32 AL cases. Results showed the bcl 2 mRNA expression of the bone marrow cells was remarkably higher than that of the control group (P<0.01). After the treatment, the bcl 2 mRNA was obviously lower than that of the control group (P>0.01), but there was no significance difference in bax mRNA in the both groups (P<0.05). The analysis of the expression ratio of bcl2 □and bax Mrna showed the nonremission (NR) rate in the high ratio group. Signifcantly higher than that in lower ratio group (P<0.01). Result shoused There is a relationship between AL and the abnormal expression of bcl 2 and bax bcl 2 and bax mRNA and their ratio may be served as a predictor for the determination of AL prognosis and treatment.
