摘 要:目的探讨胡桃醌与顺铂(DDP)联合应用对人肝癌HepG2细胞增殖的影响。方法采用不同浓度的胡桃醌或(和)顺铂处理肝癌HepG2细胞24 h,采用倒置显微镜观察细胞的形态学变化,通过MTT法检测其对HepG2细胞生长的抑制作用。结果不同浓度的药物作用可导致HepG2细胞发生形态学变化,MTT比色法测定显示,采用胡桃醌、顺铂合用,可显著抑制HepG2细胞的增殖,单用不同浓度的胡桃醌与顺铂分别作用于肝癌HepG2细胞后可以抑制其增殖且呈剂量依赖性。胡桃醌联合顺铂增强了HepG2细胞的凋亡。结论胡桃醌联合顺铂与单药相比能更显著抑制人肝癌HepG2细胞的增殖,促进其凋亡,两药对肝癌细胞的杀伤效应具有一定的协同作用。Objective To explore the effect of Juglone combined with DDP on the proliferation of HepG2 cells. Methods Different doses of Juglone (DDP) were used to treat HepG2 cells,and optical microscope was used to ob- serve the morphological changes of HepG2 cells, the proliferation inhibition of drugs on HepG2 cells was measured by MTT assay. Results Different doses of drugs lead to the morphological changes of HepG2 cells. MTT assay indicate that Juglone combined with DDP could obviously inhibit the proliferation of HepG2 cells in a dose dependent manner. Juglone combined with DDP could enhance the apoptosis of HepG2 cells. Conclusion Juglone combined with DDP could obviously inhibit the proliferation of HepG2 cells compared with different doses of Juglone or DDP alone, and en- hance the apoptosis of HepG2 cells, Juglone and DDP have the synergistic effect on the HepG2 cells apoptosis.
