摘 要:目的研究肾透明细胞癌(ccRCC)组织中CXC趋化因子受体4(CXCR4)和基质金属蛋白酶-2(MMP-2)的表达及意义。方法用免疫组织化学PV6000和RT-PCR法联合检测88例ccRCC组织和48例正常肾组织中CXCR4和MMP-2蛋白及mRNA的表达。结果在ccRCC组织中CXCR4和MMP-2蛋白及mRNA的表达显著高于正常肾组织(P<0.05);在ccRCC组织中CXCR4和MMP-2蛋白及mRNA的表达与肿瘤的临床分期、病理分级及有无淋巴结转移相关(P<0.05),而与患者年龄、性别及肿瘤直径无关;在ccRCC组织中CXCR4和MMP-2的表达在蛋白水平及基因水平均呈正相关(P<0.05)。结论 CXCR4和MMP-2可能在ccRCC的发生、发展、侵袭和转移过程中发挥协同作用,有望为ccRCC诊治提供新途径。Objective To study the expression and significance of CXC chemokine receptor 4(CXCR4) and matrix metalloproteinases-2 ( MMP-2 ) in clear cell renal cell carcinoma ( ccRCC ) tissues. Methods The expression of CXCR4 and MMP-2 mRNA in 88 cases of fresh ccRCC tissues and 48 cases of normal renal tissues were detected by RT-PCR. Meanwhile, the expression of CXCR4 and MMP-2 protein was examined by immunohistochemical PV6000 assay. Results The expression of CXCR4 and MMP-2 mRNA and protein in ccRCC tissues were signifi- cantly higher than those in normal ones(P 〈 0.05). Furthermore, the expression of CXCR4 and MMP-2 in ccRCC tissues had a significant relationship with clinical stage, pathological grade and lymph node metastasis ( P 〈 0. 05 ) , while not with gender and age of patients and the diameter of tumors. Moreover, there was a positive correlation be- tween the expression of CXCR4 and MMP-2 in ccRCC tissues ( P 〈 0.05 ). Conclusions CXCR4 and MMP-2 may have a synergistic effect on the process of occurrence,development, invasion and metastasis of ccRCC, and thus provide a novel strategy for diagnosis and therapy of ccRCC.
