摘 要:目的:了解正常人口腔黏膜全基因组甲基化状态。方法:应用甲基化DNA免疫沉淀技术检测正常人口腔黏膜全基因组甲基化。结果:正常人口腔黏膜中检测出的甲基化CpG岛占总数的16.4%,其中,位于基因启动子区段的甲基化CpG岛占40.2%,甲基化CpG岛和甲基化基因均主要集中于1、2、16、17、19号染色体,甲基化的基因主要集中在与癌症、MAPK、钙信号相关的通路中。结论:人正常口腔黏膜存在甲基化现象,甲基化CpG岛主要集中在富含基因的染色体上。Objective: To understand the genome-wide methylation status in oral mucosa of healthy people. Method: The oral mucosa of healthy subjects was examined by methylated DNA immunoprecipitation. Result: In normal oral mucosa, methylated CpG islands accounted for 16.4 % of the total CpG islands. Methylated CpG islands mainly located in promoter region (40.2 %). Methylated CpG islands and methylated genes were focused on chromosome 1,2, 16, 17 and 19. Most likely they are involved in the pathways associated with cancer, MAPK, calcium signals. Conclusion: Methylation presents in hu- man normal oral mucosa and methylated CpG islands are primarily located in the gene-rich chromosomes.
