摘  要:新霉素 (Neomycin)是由链霉菌属弗氏链霉菌 (Streptomycesfradiae)经发酵培养产生的一种氨基糖甙类抗生素 ,目前 ,它作为一种常用兽药和具有发展前景的饲料添加药物在畜牧业中广为使用。为了控制新霉素在动物性食品中的残留 ,保障食品卫生和公共健康 ,本研究首次在国内对鸡体内新霉素残留的微生物学检测方法进行了研究。本研究选择 2倍于样本重量的 0 1mol/L磷酸盐缓冲液 ( pH8 0 )为组织提取液 ,经加热、离心除去蛋白质 ,用测试菌为表皮葡萄球菌 (Staphylococcusepidermidis)的微生物学杯碟法测定提取液中新霉素的含量。本方法的灵敏度为 0 15~ 0 2 μg/mL ,检测限为 0 5~ 0 75μg·g- 1组织 ,回收率为 85 3%~ 93 2 % ,回收率比FDA的传统方法有显著提高。Neomycin is classified as a broad spectrum antibiotic that is used for preventing diseases and increasing the efficiency of feed utilization in veterinary medicine.In order to control neomycin residues in food derived from animals and ensure public health,we have studied on determination of neomycin in chicken tissues by cylinder plate assay of microbiological method. Staphylococcus epidermidis was taken as the test microorganism,neomycin in tissues(chicken muscle,liver,kidney and fat)were extracted by 0 1 mol/L potassium phosphate buffer (pH8 0) and the ratio of buffer volume to tissues weight was 2∶1,the protein in tissues was precipitated by heating.The slope of the standard curves for tissues (muscle,liver,kidney and fat)were in range of 0 0977~0 1273,with cofficients of variation of 1 1%~9 7%,the sensitivity of the method were 0 15~0 2 μg/mL.The recovery rate of neomycin from tissues fortified at 0 5~3 0 mg/kg ranged from 85 3%~93 2%,with coefficient of variation of 2 3%~8 2%,the limits of detection for neomycin in tissues were 0 5~0 75 μg/g.
