摘 要:通过对7株发酵性接合酵母菌株进行初筛和复筛得到1株富铁能力强的菌株F-13,以此菌株为出发菌,对其培养条件进行了优化。在单因素试验的基础上,对影响富铁酵母发酵的氮源、碳源、时间等因素进行考察,测定菌株F-13在不同影响因素下的生物量及铁含量,然后通过正交试验对菌株生物量及铁含量影响较大的因素水平进行进一步优化,确定富铁酵母最佳培养条件为:培养温度28℃,pH值6.0,培养基Fe2+添加浓度为120 mg/l,摇床转速200 r/min,接种量10%,装液量50 ml/250 ml三角瓶,培养时间48 h。在此优化条件下获得的富铁酵母生物量可达到3.92 g/l,铁含量可达7.04 mg/g。Through the 7 Saccharomycescerevisiae strains for primary and secondary screening, an strong iron-rich capabilities strain F-13 was screened. Taking this strain of bacteria as a starting strain, the culture conditions was optimized. First, the single factor experiment was based on the effect of iron-rich yeast nitrogen, carbon, time and other factors, strain F-13 was measured at different fac- tors affecting biomass and iron content. Then, the orthogonal experiment was made to further optimize the factor level which impacted the biomass and iron content greater. Determine the best iron-rich yeast culture conditions: culture temperature 28 ℃, pH value of 6.0, the medium concentration of Fe2+ 120 mg/l, shaking speed 200 r/min, 10% inoculum, liquid volume 50 ml/250 ml, incubation time 48 h. In optimal conditions obtained from iron-rich yeast, biomass was 3,92 g/l, iron content was up to 7.04 mg/g.
