摘 要:目的探讨雷公藤内酯对海人酸致痫大鼠海马区水通道蛋白4(AQP4)表达的影响。方法 30只SD大鼠随机分成三组:对照组、海人酸组、雷公藤内酯干预组,每组10只。海人酸组颈内皮下注射海人酸,剂量为10 mg/kg;雷公藤内酯干预组在海人酸注射前连续7 d腹腔内注射雷公藤内酯,剂量为30μg/kg;对照组注射等量的生理盐水。造模成功后取脑海马组织,应用RT-PCR和Western blot法检测各组动物海马区AQP4蛋白的表达。结果 Western blot结果显示,海人酸组和对照组AQP4条带与GAPDH条带的灰度值比分别为0.872±0.141和0.453±0.078,海人酸组AQP4表达显著增多(P<0.05);雷公藤内酯干预组中海马AQP4蛋白相对灰度值比为0.647±0.185,表达较海人酸组减少(P<0.05)。RT-PCR结果显示,海人酸组与对照组AQP4条带与GAPDH条带的灰度值比分别为1.212±0.115和0.843±0.091,海人酸组AQP4 mRNA表达比对照组显著增加(P<0.05);雷公藤内酯干预组AQP4 mRNA相对灰度值比为1.105±0.192,与海人酸组比较明显减少(P<0.05)。结论雷公藤内酯对海人酸致痫大鼠脑海马区AQP4蛋白的表达有下调作用,可以作为临床难治性癫痫的候选药物之一。Objective To investigate the effect of triptolide on the expression of AQP4 protein in hippocampus of kainite-induced epilepsia rat.Methods 30 SD rats were divided into 3 groups,each group had 10 rats,including control group,kainite-induced group and triptolide treatment group.Kainite-induced group was endothelial injected with kainic acid of 10 mg/kg.Triptolide treatment group was intraperitoneal injected with triptolide of 30 μg/kg before kainic acid injection for 7 consecutive days.Control group was injected with normal saline.Hippocampus was taken from brain after establishing the model successfully.Western blot and RT-PCR were used to examine the expression of AQP4 protein in the hippocampus of rats.Results Western blot results showed that the expression ratio of AQP4 and GAPDH in the hippocampal region of kainite-induced group was 0.872±0.141,which increased significantly than control group (0.453±0.078,P < 0.05).The expressions ratio of AQP4 and GAPDH in the hippocampal region of triptolide treat ment group was 0.647±0.185,which obviously decreased than kainite-induced group (P < 0.05).RT-PCR results showed that the expression ratio of AQP4 and GAPDH in the hippocampal region of kainite-induced group was 1.212±0.115,which increased significantly than control group (0.843±0.091,P < 0.05).The expressions ratio of AQP4 and GAPDH in the hippocampal region of triptolide treatment group was 1.105±0.192,which obviously decreased than kainite-induced group (P < 0.05).Conclusion Triptolide can inhibit the expression level of AQP4 protein in the hippocampal region in epilepsy rats and play a important role in the drug therapy.
