摘  要:目的:观察使用诱导型一氧化氮合酶抑制剂对大鼠门脉高压性胃病的影响。方法:健康雄性SD大鼠60只随机分为5组,空白组、对照组、低剂量组、中剂量组和高剂量组。空白组分离门静脉、关腹,其余组门脉缩窄法造门脉高压性胃病模型。空白组与对照组手术后腹腔注射生理盐水,其余3组手术后腹腔注射不同浓度SMT。术后14d,检测每组大鼠门脉血和胃壁组织Total Nitric Oxide Synthase(TNOS)、Inducible Nitric Oxide Synthase(iNOS)的活性及NO的浓度,免疫组化CD34标记胃血管内皮,测量每组大鼠胃黏膜下血管数目、面积。结果:对照组大鼠门脉血及胃壁组织TNOS活性、iNOS活性、NO浓度、胃黏膜下血管直径及血管总面积较空白组显著升高(P<0.01)。中、高剂量组大鼠门脉血及胃壁组织TNOS活性、iNOS活性、NO浓度、胃黏膜下血管直径及血管总面积较对照组均显著下调(P<0.05)。结论:大鼠门脉高压性胃病发病机制中有NO的参与,在门脉缩窄型门脉高压性胃病中NO主要由iNOS生成,SMT对大鼠门脉高压性胃病可能具有一定保护作用。Objective:To observe the effect of inducible nitric oxide synthase inhibitor SMT on portal hypertension gastropathy in rats.Methods:60 healthy male SD rats were randomly divided into five groups:sham operation group,model group,low dose SMT treatment group,middle dose SMT treatment group and high dose SMT treatment group.The portal vein was not ligated in sham-operated group.The other groups were given a partial ligation of the portal vein.Sham-operation group and model group were given normal saline through intraperitoneal injection.Other three groups were given different conecntrations SMJ through intraperitoneal injection.14 days after surgery,portal vein pressure,the total nitric oxide synthase(TNOS) activity,inducible nitric oxide synthase(iNOS) activity and NO content in portal vein and stomach tissue were measured.The number and area of gastric wall submucosal vessels marked by CD34 were measured by image analytical apparatus.Results:The model group have a significant increase in the TNOS activity,the iNOS activity,the NO content of portal blood and stomach tissue,as well as in the diameter and area of gastric wall submucosal vessels comparied with the sham-operation group(P〈0.01).However,the middle-dose SMT treatment group and high-dose SMT treatment group have a significant decrease in the above parameters comparied with the model group(P〈0.05).Conclusion:NO play an important role in the pathogenesis of portal hypertension gastropathy in rats,NO is major produced by iNOS in portal hypertension gastropathy induced by partial portal vein ligated.SMT probably have some protective effect on portal hypertension gastropathy in rats.
