摘 要:目的研究骨髓间充质干细胞(bone marrow stromal cells,BMSCs)移植对兔深静脉血栓的治疗作用。方法取新西兰大白兔40只,其中8只作为干细胞供体,余32只建立血栓模型,随机均分为4组。A、C组分别于建模3、10 d后,将含有107个BMSCs的DMEM悬液移植于股静脉中;B、D组分别于建模3、10 d后,将0.9%氯化钠注射液注入股静脉内。移植后14、28 d对病变段下腔静脉行HE染色及血管性血友病因子(vWF)免疫组化染色观察新生毛细血管数;移植后14 d采用Western blot法检测血栓组织中血管内皮细胞生长因子(VEGF)及碱性成纤维细胞生长因子(bFGF)蛋白的表达。结果移植后14、28 d计算每5个高倍视野新生毛细血管数,A、C组多于B、D组(P<0.05),C组多于A组(P<0.05),B组与D组比较差异无统计学意义(P>0.05)。各组VEGF、bFGF蛋白均有表达,A组VEGF、bFGF蛋白水平高于B组(P<0.05),C组高于D组(P<0.05),B、D组比较差异无统计学意义(P>0.05)。结论 BMSCs移植能明显增加兔下腔深静脉血栓段中新生毛细血管数,改善血栓机化再通情况,同时能明显增加VEGF、bFGF的分泌,两者的正反馈作用能更好地促进血栓的机化和再通。Objective To explore the effect of transplantation with bone marrow stromal cells (BMSCs) in treat- ment of deep vein thrombosis in rabbits. Methods Forty New Zealand rabbits, of whom 8 were used as stem cell donors and other 32 were randomly divided into group A, B, C and D (n = 8) after models of deep vein thrombosis were estab- lished. The 107 BMSCs of dulbecco's modified eagle medium (DMEM) suspensions were injected into femoral veins at 3th d and 10th d after modeling in group A and C; while the 0.9% sodium chloride injections were injected into femoral veins at 3th and 10th d after modeling in group B and D. The capillary density was detected with capillaries counting after hematoxylin and eosin (HE) staining and von willebrand disease factors (vWF) immunohistochemieal staining at 14th and 28th d after transplantation ; protein expressions of vascular endothelial cell growth factor ( VEGF ) and basic fibroblast growth factor (bFGF) in thrombus tissues were detected with Western Blot method 14 d after the transplantation. Results On the 14th and 28th d after transplantation, capillary numbers in each 5 high power field were significantly higher in group A and C than those in group B and D (P 〈 O. 05), and the number in group C was higher than that in group A (P 〈 0.05 ) , but there was no significant difference between group B and D (P 〉 O. 05). Expressions of VEGF and bF- GF were found in each group. Protein expressions of VEGF and hFGF in group A were significantly increased than those in group B ( P 〈 0. 05 ), and the expressions in group C were higher than those in group D ( P 〈 0.05 ), but there were no significant differences between group B and D (P 〉 0. 05). Conclusion BMSCs transplantation cas obviously increase the capillary number in rabbits'deep vein thrombosis, improve the thrombus organization recanalization and increase the secretions of VEGF and bFGF, all of which helps to accelerate organization and recanalization of vein t
