摘 要:研究通过直接测序法检测ASIP基因编码区和部分内含子区域突变位点,以确定ASIP基因突变是否影响中国绵羊的被毛表型变化.结果表明:2个缺失突变(9-bp,c.10-18,D9;5-bp,c.100-105,D5)位于ASIP基因第2外显子上,3个SNPs(g.672G>A,g.1580G>A和g.1617G>A)位于第2内含子区域;D9和D5缺失突变在10个绵羊群体中都存在,而且在‘岷县黑裘皮绵羊’和‘多浪羊’群体内各出现1只D5D5基因型个体,在所有检测个体中没有发现D9D9基因型;结合绵羊被毛表型相关分析,D9和D5缺失突变与所研究的绵羊品种被毛表型不存在相关或不完全相关,即ASIP基因编码的蛋白质区域变异不能解释中国绵羊群体毛色表型的变异.The aim of this research was to understand that if mutations from the ASIP gene affect coat phenotype variation in Chinese sheep through direct Sequencing. The results showed that two deletion mu- tations and three SNPs were identified, a 9-bp deletion (c. 10-18del) and 5-bp deletion (c. 100-105del), both of which were located in exon 2,three SNPs(g. 672 G〉A,g. 1580 G〉A 和g. 1617 G〉A)were located in intron 2. Two deletion mutations were presented in 10 Chinese sheep breeds. Moreover, only two sheep have the D5D5 genotype, one in 'Minxian black-fur sheep' and one in 'Duolang sheep', no homozygosis D9D9 was found in all detected sheep. The genotype results suggested that these mutations were not associ- ated or not completely associated with coat color in the investigated sheep breeds. These results indicated that the variation in the protein coding region of ASIP did not explain the coat color phenotypes variation of Chinese indigenous sheep breeds.
