摘  要:采用逆转录PCR(RT-PCR)及cDNA末端快速扩增(RACE)技术从虹鳟(Oncorhynchus mykiss)脑组织中克隆了Y-box基因cDNA全长序列(GenBank登录号:FJ492962)。该基因cDNA全长为1 452 bp,其中5'端非翻译区(5'UTR)长137 bp,3'端非翻译区(3'UTR)长409 bp,开放阅读框(ORF)长906 bp,编码301个氨基酸。蛋白质相对分子质量为33.1×103。PSORTⅡ亚细胞定位显示该蛋白位于细胞核,富含两类核定位信号,分别为pat4与pat7。虹鳟的Y-box蛋白序列与大西洋鲑、斑马鱼和大菱鲆的同源性分别为99%、80%和79%。同源建模显示虹鳟与人的Y-box蛋白具有十分相似的三级结构。系统发育树显示不同物种的Y-box蛋白分为3个亚群:亚群Ⅰ为鱼类;亚群Ⅱ为两栖类、鸟类及哺乳类;亚群Ⅲ为节肢动物类,这与物种间的进化历程基本一致。Using reverse transcription PCR (RT-PCR)and rapid amplification of cDNA ends (RACE)technique, a full-length sequence of Y-box gene of cDNA( GenBank No. : FJ492962 )from rainbow trout (Oncorhynchus mykiss )brain tissue was cloned. The full-length eDNA was 1 452 bp, with 5' untranslated region( 5' UTR) 137 bp,3' untranslated region ( 3' UTR) 409 bp, open reading frame (ORF)906 bp, encoding 301 amino acids. The molecular weight is 33.1 × 10^3. PSORT Ⅱ subeellular localization showed that the protein was in the nucleus, which contains two kinds of nuclear localization signal of pat4 and pat7. The Y-box protein sequences homology among rainbow trout, zebrafish, Atlantic salmon and turbot were 99% , 80% and 79%. Homology modeling shows rainbow trout and human Y-box proteins had the similar tertiary structure. Phylogenetic tree showed that different species of Y-box proteins were divided into three subgroups:subgroup I for fish;subgroup Ⅱ for amphibians, birds and mammals;subgroup III for arthropod animal, which is consistent with the evolution of species
