摘 要:目的:观察珍珠水解液对正常皮肤及瘢痕皮肤来源成纤维细胞碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)、转化生长因子β1(transforming growth factor beta 1,TGF-β1)的影响,并探讨其对病理性瘢痕的作用。方法:原代培养、分离纯化获得人类皮肤瘢痕和正常皮肤成纤维细胞系。采用荧光实时定量RT-PCR及免疫细胞化学法分别检测成纤维细胞在珍珠水解液作用下bFGF、TGF-β1mRNA及蛋白表达变化的情况。结果:珍珠水解液可下调增生性瘢痕及正常皮肤来源的成纤维细胞TGF-β1mRNA和蛋白表达,同时上调bFGF的mRNA和蛋白的表达,且呈现浓度依赖性。结论:珍珠水解液能调节成纤维细胞bFGF、TGF-β1的分泌,其对皮肤瘢痕作用的功效及机制值得进一步研究。Objective:To observe the effects of pearl hydrolyzate on the expression of basic fibroblast growth factor (bFGF) and transforming growth factor beta 1 (TGF-β1) in fibroblasts derived from hyperolastic scar and normal skin,and to explore the relevant mechanism.Methods:Fibroblasts were cultured and purified from normal skin and hypertrophy scar tissue,the mRNA levels of bFGF and TGF-β1 in fibroblasts were determined by reverse transcription-fliorescence quantitative-polymerase chain reaction,and the protein levels of bFGF and TGF-β1 in fibroblasts were determined by immunocytochemistry after treated by pearl hydrolyzate.Results:Real-time RT-PCR and immunocytochemistry results showed that the treatment caused increase trend in the mRNA and the protein levels of bFGF for hypertrophic scar fibroblast cell,and also caused decrease in the human mRNA and the human protein substance level of TGF-β1.Conclusion:Pearl hydrolyzate can regulate the secretion of bFGF and TGF-β1 in derived normal skin and hypertrophy scar fibroblasts.The therapeutic effect of pearl hydrolyzate to the hypertrophic scar is worthy of further study.
