妊娠滋养细胞肿瘤组织中cyclin D1、Rb蛋白产物的表达

摘　　要：目的 :探讨增殖相关基因cyclinD1、Rb在妊娠滋养细胞肿瘤中的表达变化。 方法 :采用免疫组化S P法 ,检测 2 0例葡萄胎、15例侵蚀性葡萄胎、15例绒毛膜癌组织中两种基因蛋白产物的表达。结果 :在恶性滋养细胞肿瘤组织中 ,cyclinD1的阳性表达率随临床期别的增高呈递增趋势 ,而Rb呈递减趋势 ,二者在Ⅲ期的阳性表达率与Ⅰ期及Ⅱ期相比差异均具有显著性(P <0 0 5 ) ;化疗可降低cyclinD1在恶性滋养细胞肿瘤组织中的阳性表达 ,而对Rb无影响。 结论 :cyclinD1的过表达 。Purpose To detect the expression of the related genes of cell proliferation such as cyclin D1,Rb in gestational tropoblastic tumors. Methods Two kinds of gene product were examined in 20 complete hydatidiform moles, 30 gestational tropoblastic tumor in paraffin embedded tissues, using the streptavidin peroxidase immunohistochemical staining. Results cyclin D1 prooncogene expression increased with advancing clinical stages, however, Rb showed decreased. Cyclin D1 and Rb expression in stage Ⅲ was significantly higher than that in stage Ⅰand Ⅱ ( P <0 05, respectively). Expression of cyclin D1 can be reduced by chemotherapy, but there was no influence on the expression of Rb. Conclusions These data suggest that the overexpression of cyclin D1 and deletion of Rb antioncogene may have an important biological significance in trophoblastic tumors.
