摘 要:目的研究血红素加氧酶-1(HO-1)对高糖作用下体外培养的大鼠腹膜间皮细胞(RPMC)转化生长因子β1(TGF-β1)和纤维连接蛋白(FN)表达的影响。方法将原代培养的第3代RPMC随机分为对照组、高糖组、钴原卟啉(CoPP)+高糖组和CoPP组。同步培养72h后,以反转录聚合酶链反应(RT-PCR)法检测各组细胞TGF-β1和FNmRNA表达情况;酶联免疫吸附法(ELISA)检测细胞上清液中TGF-β1和FN的蛋白质水平。结果高糖组、CoPP+高糖组和CoPP组的HO-1mRNA及蛋白表达水平均显著高于对照组,其中CoPP+高糖组显著高于高糖组;高糖组和CoPP+高糖组的FNmRNA及蛋白表达水平显著高于对照组及CoPP组;高糖组、CoPP+高糖组TGF-β1mRNA及蛋白表达水平显著高于对照组及CoPP组,其中CoPP+高糖组显著低于高糖组。结论 HO-1可抑制高糖诱导的RPMCTGF-β1和FN的表达,具有抗腹膜纤维化的作用。Objective To explore the influence of heme oxygenase-1(HO-1)on the expressions of transforming growth factor-β1(TGF-β1)and fibronectin(FN)in peritoneal mesothelial cells(RPMC)of rats induced high glucose.Methods The third generation of cultured RPMC were randomly divided into control group, high-glucose group, cobalt protoporphyrin(CoPP)+ high-glucose group and CoPP group.After spontaneous cultivation for 72 hours, reverse transcription-polymerase chain reaction(RT-PCR)was used to detect the expressions of TGF-β1and FN mRNA in each group, and enzyme-linked immuno sorbent assay(ELISA)was used to detect the levels of TGF-β1and FN proteins in the cell supernatant.Results The levels of HO-1 mRNA and protein expressions in high-glucose group, CoPP+ high-glucose group and CoPP group were significantly higher than those in the control group, and indexes mentioned above in the CoPP+ high-glucose group was significantly higher than the high-glucose group. The levels of FN mRNA and protein expressions in the high-glucose group and CoPP+ high-glucose group were significantly higher than the control group and CoPP group. The levels of TGF-β1mRNA and protein expressions in the high-glucose group and CoPP+ high-glucose group were significantly higher than the control group and CoPP group, and indexes mentioned above in the CoPP+ high-glucose group weresignificantly lower than the high-glucose group. Conclusion HO-1 can inhibit the expression of TGF-β1and FN in RPMC induced by high glucose, and it has the function of anti-peritoneal fibrosis.
