摘 要:目的观察环氧合酶2(COX-2)选择性抑制剂塞来昔布对人乳腺癌MCF-7细胞株和宫颈癌HeLa细胞株的放射增敏作用。方法用四唑盐比色法(MTT)检测塞来昔布对人乳腺癌MCF-7细胞株和宫颈癌HeLa细胞株的增殖抑制情况;克隆形成实验检测放射增敏作用。结果 MTT检测结果显示塞来昔布对人乳腺癌MCF-7细胞株和宫颈癌HeLa细胞株的增殖有抑制作用,呈现出浓度-时间依赖性;克隆形成实验提示塞来昔布对人乳腺癌MCF-7细胞株和宫颈癌HeLa细胞株有一定程度的放疗增敏效应。结论塞来昔布能增强人乳腺癌MCF-7细胞株和宫颈癌HeLa细胞株的放疗敏感性。Objective To observe the radiosensitization effect of celecoXib (a selective in- hibitor of cyclooxygenase-2) on human breast cancer MCF-7 cell lines and human cervical cancer HeLa cell lines. Methods The growth inhibition effects of celecoxib on human breast cancer MCF -7 cell lines and human cervical cancer HeLa cell lines were detected by MTT experiment, and the radiosensitization effect of celecoxib was detected by Cloning formation experiment. Results MTT experiment showed the growth inhibition effects of celecoxib on human breast cancer MCF-7 cell lines and human cervical cancer HeLa cell lines were expressed in a concentration and time dependent manner. Cloning formation experiment suggested that celecoxib had a certain degree of radiosensiti- zation effect on human breast cancer MCF-7 cell lines and human cervical cancer HeLa cell lines. Conclusion Celecoxib can enhance the sensitivity of radiotherapy on human breast cancer MCF-7 cell lines and human cervical cancer HeLa cell lines.
