摘  要:目的 探索伏隔核(NAc)深部电刺激(DBS)对锂一匹鲁卡品大鼠模型急性期癫痫发作的影响。方法实验大鼠按随机数字表顺序分为五组,DBS壳部组(A),DBS核部组(B),假处理壳部组(C),假处理核部组(D),对照组(E),每组6只大鼠。DBS组置入电极并给予高频率电刺激,假处理组置入电极不给予电刺激。对照组不置入刺激电极。电极置入1周后,大鼠腹腔注射锂一匹鲁卡品诱导癫痫发作。锂一匹鲁卡品给药后,马上给予DBS组大鼠高频率电刺激(130Hz,100μs,150μA)。观察大鼠行为发作强度和脑电图变化。结果高频率电刺激NAc壳部明显延长首次癫痫发作潜伏期(P〈0.05),减轻癫痫发作行为强度(P〈0.05),同时缩短第二阶段发作时程和总发作时程(P〈0.05)。而电刺激NAc核部对癫痫发作无明显改善。结论高频率电刺激NAc壳部明显抑制匹鲁卡品诱导的癫痫发作。Objective To explore the effect of high frequency deep brain stimulation (DBS) to nucleus accumben (NAc) on an experimental seizure model induced by lithium - pilocarpine during the acute phase. Methods 30 rats were randomly even divided into 5 groups. In the DBS groups, the rats underwent implantation of DBS electrodes in the shell or core of NAc. Two sham - treated groups received implants but no stimulation. Last control group received no DBS electrodes. One week after these procedures, the animals were provided lithium - pilocarpine to induce seizures. DBS groups were received high frequency stimulation (130 Hz, 100 μs, 150 μA) immediately after pilocarpine administration, and changes of the magnitude of seizure and electroencephalography (EEG) were evaluated. Results Our study showed that high frequency stimulation on shell of NAc significantly prolonged the latency to develop seizure ( P 〈 0. 05 ), suppressed the severity of seizures ( P 〈 0. 05 ), and reduced the second stage and total duration of seizures ( P 〈 0. 05). However, high frequency stimulation on core of NAc failed to show any significance in inhibiting seizures. Conclusions ANAc shell high frequency stimulation was protective against seizures induced by pilocarpine.
