摘  要:目的:探讨人类生物钟基因hClock及hBmal1蛋白在人类胆管癌组织中的表达,并研究其表达与人类胆管癌的关系。方法:采用免疫组织化学法检测60例人类胆管癌组织及癌旁组织中hClock基因、hBmal1基因蛋白产物(Clock蛋白及Bmal1蛋白)的表达。结果:60例人胆管癌组织与相应癌旁组织中,Clock蛋白在胆管组织中阳性率为33.33%(20/60),在癌旁组织中阳性率为81.67%(49/60),两者阳性表达率差异有统计学意义(P<0.05);而Bmal1蛋白在胆管癌组织中阳性率为73.33%(44/60),癌旁组织中阳性率为36.67%(22/60),两者阳性表达率差异也有统计学意义(P<0.05)。Clock蛋白与Bmal1蛋白表达均与胆管癌TNM分期有关(P<0.05)。在胆管癌组织中Clock蛋白与Bmal1蛋白表达呈负相关(r=-0.481,P<0.05)。结论:Bmal1蛋白可能与人类胆管癌的发生有关,与侵袭、转移的关系有待深入研究,而Clock蛋白可能为胆管癌发生的抑制因素。Objective- To investigate the expression of human circadiancolock genes hCIock protein and hBmall protein in chotangiocarcinoma tissues and its cooretation with cholangiocarcino- ma invasion and metastasis. Methodst The expressions of hCIock and hBmall genes protein prod-uct (Clock protein and BMALt protein) in 60 cases of cholangiocarcinoma and para-neoplastic tis- sues were detected by immunohistochemistry method. Results.. In 60 cases of cholangiocarcino- ma tissues Clock protein positive expression rate was 33.33% (20/60), the Clock protein of para-neoplastic tissues positive expression rate was 81.67% 49/60), there was significant differ- ence between them(P〈0.05). The Bmall protein of cholangiocarcinoma and para-neopastic tissues positive expression rate were 73.33% (44/60) and 36.67% (22/60), there was significant difference between them P〈0.05). The Clock protein and Bmall protein had significant relationship with TNM clinical stage (P〈0.05). The Clock protein and BMAL1 protein in cholangiocarcinoma tissues were negative correlation (r=-0.481, P〈0.05). Conclusion: The BMAL1 protein and the occurrence of cholangiocarcinoma are related,but the relationship wiith invasion and metastasis need to be studied in greater depth,and the Clock protein may be one of the inhibition factors of the occurrence of cholangiocarcinoma.
