摘 要:目的 探讨人泪腺腺样囊性癌(LACC)细胞系裸鼠异种种植模型的建立及荷瘤的组织生物学行为.方法 实验研究.将人LACC细胞系接种于10只裸鼠腋下的皮下,注射细胞悬液后每天观察裸鼠以及测量裸鼠荷瘤的最长径L和最短径W,并用公式V(荷瘤体积)=(LxW2)/2计算出荷瘤的体积.分别于注射细胞悬液后14、28、35、42、49 d随机取2只裸鼠的荷瘤以及裸鼠的肝脏、肺和腋下淋巴结.对注射细胞后14 d的荷瘤行透射电镜观察,对注射细胞后14、28、35、42、49 d的荷瘤行HE染色、免疫组织化学染色检测Keratin、Vimentin、α-SMA、S-100、Desmin的表达以及提取其总RNA,用RT-PCR检测该荷瘤的人类β-actin的表达情况.对所取的裸鼠肝脏、肺和腋下淋巴结组织切片,行HE染色,观察有无肿瘤转移.结果 接种细胞系后7d,10只裸鼠腋下皮下全部长出荷瘤.HE染色和透射电子显微镜下观察,荷瘤具有典型的上皮性肿瘤组织结构.对荷瘤行免疫组织化学染色示Keratin、Vimentin、α-SMA、S-100阳性,Desmin阴性.RT-PCR检测荷瘤表达人类β-actin基因.光镜下观察,均未发现肿瘤转移至裸鼠肝脏、肺和腋下淋巴结.结论 人LACC细胞具有一般恶性肿瘤的特点,同时也具有腺样囊性癌细胞的特点.荷瘤具有与原瘤一致的组织学特性,该动物模型易复制,时间短,成功率高,是研究LACC的良好模型.Objective To investigate characterization of human lacrimal adenoid cystic carcinoma line LACC and establish a model of human LACC.Methods Experimental research.Human LACC cells were injected into subcutaneous tissue of nude mice.The length (L) and width (W) of the tumor were measured every day after injection of LACC cells,and the tumor volume was calculated as L × W2 × 1/2.Two mice bearing LACC tumor were randomly killed,removed the tumors anatomy and their lungs,livers,and lymph nodes of oxter,14,28,35,42,49 days after injection of LACC cells.Histologic examination,immunohistochemical analysis for Keratin,Vimentin,α-SMA,S-100,Desmin,PCR analysis for human β-actin and electron microscopy were performed.Whether tumor metastasis to lungs,livers and lymph nodes of oxter were observed by hematoxylin-cosin staining method.Results Heterotransplanted tumors were observed in all 10 mice after injection of LACC cells.The results of HE staining and transmission electron microscope indicated that heterotransplanted tumor possesses typical histological characterization of epithelial carcinoma.The results of immunohistochemistry showed that keratin,S-100,Vimentin,α-SMA expression in tumor tissue were positive,but Desmin expression were negative.RT-PCR analysis revealed that tumors expressed human β-actin,indicating their human origin.Histologic examination show that tumors didn't metastasize to lung,liver and lymph node.Conclusions Human LACC cell line possesses characterization of malignant carcinoma and also possesses characterization of adenoid cystic carcinoma.It is found that the heterotransplanted LACC tumor has histologic features similar to the original LACC of the patient.This model with lacrimal cystic adenoid carcinoma in nude mice is relatively easy,quick,and especially with high successful rate.So it can be considered as an ideal animal model for study on lacrimal cystic adenoid carcinoma in vivo.
