microRNA-195在食管鳞状上皮细胞癌中的表达及临床意义

摘　　要：目的检测microRNA-195(miR-195)在食管鳞状上皮细胞癌组织中的表达水平,探讨其临床病理生理学意义。方法应用实时荧光聚合酶链反应法检测94例食管鳞状上皮细胞癌组织和配对癌旁正常组织中miR-195的表达水平,并分析miR-195表达水平与食管鳞状上皮细胞癌患者临床病理学特征及预后之间的关系。结果食管鳞状上皮细胞癌组织中miR-195不同程度低表达45例(47.9%),正常表达39例(41.5%),高表达10例(10.6%)。miR-195低表达者比例在有无淋巴结转移、不同TNM分期者中的差异有统计学意义(P<0 05);miR-195表达水平与患者无瘤生存期显著相关,是食管鳞状上皮细胞癌患者预后的独立危险因素。结论miR-195下调影响食管鳞状上皮细胞癌进展和患者预后,可能起着抑癌基因功能。Objective To investigate the expression of miR- 195 in esophageal squamous cellcarcinoma （ESCC） and its clinicopathological significance. Methods The expression of miR- 195 in 94 ESCC samples and matched normal tissue samples was detected by real- time PCR. Results 47.9%（45/94） of carcinoma samples showed low expression of miR- 195, 41.5%（39/94） was negatively expressed and 10.6%（10/94） showed high expression.The low expression of miR- 195 was significantly correlated with lymph node metastasis, TNM staging and disease free survival. miR- 195 expression level was an independent prognostic factor in patient with ESCC. Conclusion The down- regulation of miR- 195 is associated with the progression of ESCC and prognosis of the patients.
