摘 要:目的本实验模拟2型糖尿病病程中葡萄糖、胰岛素的浓度改变,研究不同浓度的葡萄糖、胰岛素对SW480肿瘤细胞分泌IGF-1的影响。方法体外培养SW480肿瘤细胞。依据培养液中加入葡萄糖、胰岛素浓度的不同,将实验细胞分为六组:0组(对照组):RPMI 1640细胞培养液;第1组:RPMI1640细胞培养液+5 mmol/L葡萄糖和5μIU/ml胰岛素;第2组:RPMI 1640细胞培养液+5 mmol/L葡萄糖和125μIU/ml胰岛素;第3组:RPMI 1640细胞培养液+25 mmol/L葡萄糖和125μIU/ml胰岛素;第4组:RPMI 1640细胞培养液+25 mmol/L葡萄糖和5μIU/ml胰岛素;第5组:RPMI 1640细胞培养液+25 mmol/L葡萄糖和1.25μIU/ml胰岛素。每个组设3个复孔,于37℃、5%CO2饱和湿度的培养箱中培养48 h,培养结束后,收集细胞培养上清,用ELISA方法检测IGF-1的浓度。结果在高浓度的胰岛素组(第2组、第3组,培养液中均含有125μIU/ml的胰岛素)IGF-1的浓度[(286.85±23.51)μg/L、(293.43±30.57)μg/L]高于对照组0组[(197.99±22.50)μg/L],差异有统计学意义(F=9.726,P<0.01);而第1、4、5组(培养液中分别含有5μIU/ml、5μIU/ml、1.25μIU/ml的胰岛素)IGF-l的浓度[(203.22±28.89)μg/L、(209.47±25.77)μg/L、(195.33±20.88μg/L)]与对照组0组相比较,差异均无统计学意义(P>0.05)。结论高浓度的胰岛素能够上调SW480肿瘤细胞IGF-1的表达。Objective To study the effect of different concentrations of glucose and insulin on the expression of IGF-1 in SW480 cancer cells by imitating the change of glucose and insulin in the course of type 2 diabetes mellitus. Methods SW480 cancer cells were cultured in vitro. Based on the concentrations of glucose and insulin in cell culture medium, cells were divided into six groups: group 0 (control): RPMI 1640; group 1:RPMI1 640+5 mmol/L glucose and 5uIU/ml insulin; group 2: RPMI 1640+5 mmol/L glucose and 125 uIU/ml insulin; group 3: RPMI1640+25 mmol/L glucose and 125 uIU/ml insulin; group 4:RPMI1640+25 mmol/L glucose and 5 uIU/ml insulin; group 5: RPMI 1640+25 mmol/L glucose and 1.25 uIU/ml insulin. After put in different cell culture medium, cells were cultured for 48 hours, and the supernatant were gathered. The concentrations of IGF-1 were tested by enzyme-linked immunosorbent assay (ELISA). Results In the high concentration of insulin groups (group 2 and group 3, 125 uIU/ml insulin in both culture medium), the expressions of IGF-1 were higher significantly than that of group 0 [(286.85± 23.51) ug/L, (293.43±30.57) ug/L vs (197.99±22.50) ug/L, F=9.726, P〈0.01). In group 1, 4, 5, (5 uIU/ml, 5 uIU/ml, 1.25 uIU/ml insulin in culture medium, respectively), there was no statistically significant difference compared to group 0 [(203.22±28.89) ug/L, (209.47±225.77) ug/L, (195.33±20.88) ug/L vs (197.99±22.50) ug/L, P〉0.05). Conclusion High concentration of insulin may increase the expression of IGF-1 in SW480 cancer cells.
