摘  要:目的:观察大鼠脑出血后血肿周围组织48 h、72 h后ERK1/2、c-fos、c-jun的表达,探讨新清开灵注射液干预大鼠脑出血后MMP-9表达从而抑制血肿周围脑水肿的作用途径。方法:采用尾状核注射胶原酶的方法复制大鼠脑出血的动物模型,并给予新清开灵注射液进行干预,Western blot方法对脑组织ERK1/2、c-fos、c-jun进行检测。结果:脑出血后模型组c-jun各时间点表达水平均升高,72 h达到高峰,皆有统计学意义(P<0.01)。与脑出血48 h模型组相比较,同一时间点新清开灵小剂量组血肿周围脑组织c-jun表达减少,在统计学上有显著性差异(P<0.01)。与脑出血模型72 h组比较,同一时间点新清开灵小剂量组血肿周围脑组织c-jun表达减少,具有统计学意义(P<0.05),大剂量组血肿周围脑组织中c-jun表达亦减少,在统计学上有显著性差异(P<0.01)。脑出血模型组ERK1/2、c-fos各时间点表达水平均升高,48 h达高峰,但均无统计学意义(P>0.05)。与同一时间点模型组相比较,新清开灵小剂量组、大剂量组血肿周围脑组织ERK1/2、c-fos表达均降低,但在统计学上均无差异(P>0.05)。结论:通过抑制c-jun干预MMP-9表达是新清开灵注射液干预脑出血后MMP-9表达,并以进一步减轻脑水肿的可能途径。Objective:To observe the effects of New Qingkailing Injection on the expressions of ERK1/2, c -fos and c -jun following intracerebral hemorrhage. Methods : VII2 type collagenase was used for inducing rat hematencephalon. The contents of ERK1/2, c - fos and c - jun were detected by Western blot. Results :The content of c - jun was increased at 48 h and 72 h post - ICH ,and there was obvious statistical difference between the model group and normal group (P 〈 0. 01 ), however, the maximum was observed in the model of 72 h. At 48 h post - ICH, the group of new Qingkailing Injection with small - dosage can reduce the contents of c - jun ( P 〈 0. 01 ). At 72 h post - ICH, both groups of new Qingkailing Injection can reduce the contents of c - jun ( P 〈 0. 05 ) ,while the group of new Qingkailing Injection with large -dosage had more obvious effect( P 〈 0. 01 ). The contents of ERK1/2 and c -fos were increased generally in the model of 48 h and 72 h post -ICH ,while the difference between each of them and normal one was none of statisticalsignificance ( P 〉 0.05 ). Compared with the model group with the corresponding time point, both groups of new Qingkailing Injection can reduce the contents of ERK1/2 and c - fos, while the effect was not obvious (P 〉 0. 05). Conclusion : To reduce the expression of c - jun so as to regulate MMP - 9 can be the important mechanism of New Qingkailing Injection alleviating edema following ICH.
