P62、PCNA在结肠癌中的表达及其与抗核抗体的相关性研究

摘　　要：目的探讨P62、增殖细胞核抗原(PCNA)在结肠癌中的表达及其与血清抗核抗体(ANA)的相关性,评估其在结肠癌早期诊断、恶性程度及预后评价中的应用价值。方法采用免疫组化方法检测64例结肠癌组织及42例结肠腺瘤组织P62、PCNA的表达情况;用ELISA和Western blot检测患者血清P62抗体水平;血清ANA水平采用间接免疫荧光方法检测。结果结肠癌患者血清P62抗体阳性率为23.4%,血清ANA的阳性率为82.8%,均高于结肠腺瘤患者(P<0.05);64例患者的结肠癌组织标本中有48例出现P62阳性表达,而PCNA在所有结肠癌患者的组织中均可见阳性表达。结论 P62、PCNA和ANA三种指标的联合检测对鉴别结肠癌的良、恶性鉴别,结肠癌早期诊断,恶性程度及预后评估均有重要参考意义。Objective By observing the expression of P62,proliferating cell nuclear antigen（PCNA） in colon cancer and the ser-um level of autoantibody and ANA to assess their value in early diagnosis, malignancy determine, prognosis of colon cancer. Methods In this study autoantibody iesponses to P62 were evaluated by ELISA and Western blot. Indirect immunofluorescence assay in se-rum from patients with colon cancer was used to detect the ANA level. Immunohistochemistry（IHC） study with tissue array slides was also performed to analyze protein expression of P62 and PCNA in colon cancer and. Results The result showed that P62 had a relative higher frequency of autoantibody response in colon cancer patients（23.4%） compared with patients with adenoma of colon （4.8%）. Of 64 colon cancer tissue specimens examined,48 specimens（75. 0%） expressed P62 and 64 specimens（100. 0%） ex-pressed PCNA. The ANA positive rate of colon cancer patients was 82.8%, higher than patients with adenoma of colon （21.4%） significantly（P〈0.05）. Conclusion The data suggested that the combination of P62,PCNA and ANA tests are valuable for the early diagnosis, malignancy determine, prognosis of colon cancer.
