摘  要:目的:采用反相HPLC建立何首乌提取液益生菌发酵前后指纹图谱,观察何首乌提取液发酵前后化学成分变化与活性的相关性。方法:以Agilent ZORBAX SB-AQ(4.6 mm×150 mm,5.0μm)为色谱柱,以A(乙腈)和B(0.1%磷酸)为流动相梯度洗脱,流速0.8 mL·min-1,柱温20℃,检测波长254 nm。结果:建立了有13个共有峰的HPLC色谱图,并比较了色谱峰相对峰面积的变化。结论:何首乌提取液经益生菌发酵后化学成分的变化较大,中药经过发酵可通过多途径影响化学成分,进而改变药物的药理药效。Objective:To establish chromatographic fingerprints of Polygonum multiflorum extract before and after fermentation with the reversed-phase HPLC ,and to find the correlation between chemical compo- nents and activities. Methods:The chromatographic fingerprints were obtained on an Agilent ZOR BAX SB-AQ(4.6mm× 150mm,5.0 μm) with the gradient elution solvent system of A(acetonitrile) and B(0.1% phosphoric acid). The flow rate was 0.8 mL.min-1,the column temperature was maintained at 20 ℃ and the detection wavelength was set at 254 nm. Results:Establishing HPLC prints that contain 13 common peaks on the basis of determination of the samples of Polygonum multiflorum extract before and after probiotics' fermentation ,and comparing the changes of the relative peak area. Conclusion:There is great difference be- tween the impacts of probiotics' fermentation to different components of Polygonum multiflorum extract be- fore and after fermentation. Chinese medicinal fermentation can effect chemical components through many ways and change the pharmacology and efficacy of drugs.
