摘 要:目的通过星点设计-响应面法优化石榴皮多糖的提取工艺及抗氧化活性。方法以石榴皮多糖的含量为指标,采用超声波辅助技术提取石榴皮多糖,用星点设计-响应面法研究了液料比、提取时间、提取温度和超声功率对提取石榴皮多糖的影响;观察了对二苯基苦基苯肼自由基(·DPPH)、羟基自由基(·OH)、过氧化氢(H2O2)的清除作用。结果超声波提取石榴皮多糖的最佳工艺为:液料比29.98(mL∶g),提取时间43.72 min,提取温度62.79℃,超声功率454.30 W,以此工艺条件得多糖提取率为7.03%;石榴皮多糖对·DPPH、·OH、H2O2有明显的清除作用。结论星点设计-响应面法用于优化提取工艺具有简便合理、稳定的特点,在优化石榴皮多糖提取条件中应用效果良好;石榴皮多糖有明显的抗氧化作用。Objective Central composite design(CCD) and response surface methodology(RSM) was used to optimize the extraction conditions of polysaccharides from pomegranate bark, its antioxidant activity was investigated. Methods The contents of polysaccharides in pomegranate bark was used as index, and the ultrasound assistive technology was used to extract the polysaecharides in pomegranate bark. CCD-RSM was used to study the effect of ratio of water to raw material, extraction time, extraction temperature, and ultrasonic power on extraction of polysaceharides in pomegranate bark. Antioxidant activity of polysaccharides from pomegranate bark to oxidants such as · DPPH, · OH, H2 02 was studied. Results The optimum extraction conditions were found to be ratio of water to raw material 29.98 (mL : g) , extraction time 43.72 rain, extraction temperature 62. 79 ℃, and ultrasonic power 454.30 W. Under these conditions, the extraction rate of polysaccharides was 7.03 %. Polysaecharides from pomegranate bark has obviously clearing action of · DPPH,· OH and H202. Conclusion CCD-RSM is used to optimize the extraction conditions of polysaccharides from pomegranate peel is simple, rational and stable. Polysaccharides from pomegranate bark has significant antioxidant effect.
