摘 要:目的探讨姜黄素在体外对雄激素非依赖性前列腺癌细胞PC-3生物活性的影响。方法分别采用细胞增殖与活性检测试剂盒(CCK-8)比色法、流式细胞分析、人工基底膜穿透法分析姜黄素对PC-3细胞增殖、凋亡和侵袭的影响。采用Western blot法检测姜黄素在体外对核因子κB(NF-κB)、蛋白激酶(Akt)、尿激酶型纤溶酶原激活物(uPA)蛋白表达的影响。结果姜黄素干预后,PC-3细胞的增殖水平和细胞迁移率下降,而凋亡率提高(P<0.01)。姜黄素能在体外抑制PC-3细胞的NF-κB、Akt、uPA蛋白的表达(P<0.01)。结论姜黄素可在体外显著抑制雄激素非依赖性前列腺癌细胞PC-3的增殖和侵袭,并诱导细胞凋亡;其机制可能通过抑制NF-κB、Akt、uPA等信号通路。Objective To study the inhibitory effect of curcumin on androgen-independent prostate cancer cells PC-3 in vitro. Methods The effects of curcumin on the proliferation, apostosis and invasion of PC-3 cells wer'e determined by cell counting kit(CCK-8), Annexin V/PI and matrigel invasion assays, respectively. The protein expressions of NF-κB, Akt and uPA were detected by Western blot analysis. Results After curcurnin intervention, the proliferation and invasion of PC-3 cells were significantly inhibited, apostosis rate of PC-3 cells was significantly promoted dose- and time dependently (P〈0. 01). The expressions of NF-κB, Akt and uPA were inhibited significantly (P〈0.01). Conclusion Curcumin can inhibit the proliferation and invasion, and promote the apostosis of androgen-independent prostate cancer cells PC-3 in vitro,which is probably related with the inhition of NF-κB,Akt and uPA.
