摘  要:目的 探讨IGF2R基因多态性与胃癌易感性的关系.方法 提取华北地区105例胃癌患者及235例健康查体者外周血基因组DNA,用聚合酶链反应-限制性片段长度多态性(polymerase chain reation-restriction fragment length polymorphism,PCR-RFLP)方法检测IGF2R codon 901和codon 2020位点的基因型分布及差异.结果病例组codon 2020基因型AG、GG的分布频率显著低于对照组,且携基因型AG、GG者胃癌发病风险分别是携AA者的0.65倍(95% CI:0.38~0.89,P<0.05)和0.72倍(95% CI:0.45~0.91,P<0.05).将AG、GG基因型合并计算,则其胃癌发病风险是AA基因型的0.68倍(95% CI:0.45~0.91,P<0.05).Codon 901多态性在两组间分布差异无统计学意义(P>0.05).结论 IGF2R基因codon 2020位点多态性可能对中国人群罹患胃癌具有保护作用.Objective: To evaluate the correlations of polymorphisms of IGF2R gene with individal susceptibility to gas- tric cancer. Methods : Genomie DNA were extracted from peripheral blood leukocytes of 105 gastrie cancer patients and 235 healthy controls. C, enotypes at eodon 901 and eodon 2020 polymorphie sites were identified by polymerase chain reaction- restriction fragment length polymorphism(PCR-RFLP). Results:At eodon 2020, the frequency of AG or GG genotype was higher in the gastrie eaneer patients than in the healthy controls. As compared with AA genotype, AG, GG had significantly decreased risks of gastric cancer, with the decrease being 0.65-fold ( ( 95 % CI : 0.38 ~ O. 89, P 〈 0.05 ), 0.72-fold ( 95 % CI:0.45 ~ 0.91, P 〈 0.05 ), respectively. The AG, GG genotypes eombined, their risk of gastric caneer is 0.68-fold com- pared with AA genotype ( 95 % CI : 0.45 - 0.91, P 〈 0.05 ). There was no significant differenee of eodon 901 polymorphism between the patients and the controls (P 〉 0. 05). Conclusion: The polymorphism of IGF2R at eodon 2020 might be poten- tially one of the proteetive factors against gastric cancer in Chinese population.
