摘  要:目的研究肢体远端缺血预处理(limb remote ischemic preconditioning,LRP)延缓大鼠脑缺血模型DWI图像上病灶出现的机制。方法 54只成年SD大鼠随机分为3组,脑缺血模型组24只、LRP组24只和假手术组6只。用Western blot检测和免疫荧光法分别检测不同时间点(缺血后1 h、3 h、12 h、24 h)AQP4的表达情况。并通过共染对AQP4进行初步定位。结果 Western blot表明AQP4在缺血后表达下降,而LRP组的AQP4表达水平上升。免疫荧光结果与Western blot一致,AQP4阳性表达量均在缺血组降低,而LRP组的AQP4表达量增多。共染结果显示AQP4与GFAP出现共染,证明AQP4在星形胶质细胞上的表达;AQP4与CD31少量共染,提示AQP4在血管内皮细胞上的少量表达。AQP4与MAP-2未显示出共染。推测LRP组增多的AQP4主要表达在邻近血管周围的星形胶质细胞膜上及血管内皮细胞上。结论 LRP对DWI上病灶的面积延缓作用很可能通过上调AQP4实现。Objective Our previous research have shown the present of lesion in DWI was prior to limb remote is- chemic preconditioning( LRP) in ischemic models,this study was intended to further investigate the probable mechanism. Method 54 adult SD male rats were subjected to either remote preconditioning (LRP) or sham surgery and then subse- quently underwent 1 h right middle cerebral artery occlusion (MCAo). Immunofluorescence staining and Western blot were used to investigate the expression of AQP4. Result Western blot and immunostaining analysis showed that LRP upregulat- ed protein expression of AQP4 which were decreased after stroke. Conclusions LRP may inhibit edema formation caused by ischemia by upregulating the expression of AQP4.
