摘  要:目的:探讨甲状腺癌不同证型间的血清蛋白表达质谱的差异并构建诊断模型.方法:应用表面增强激光解析电离飞行时间质谱(SELDI-TOF-MS)技术分别检测19例肝郁气滞型和21痰瘀互结型甲状腺癌的血清蛋白质指纹图谱,筛选出两组间特异性表达差异蛋白,并结合生物信息学方法建立诊断模型.结果:在2000~ 20000Da范围内共检测到86个蛋白质峰,其中22个蛋白质峰在两组之间表达有显著性差异(P<0.01),17个蛋白质峰在肝都气滞组中高表达,5个蛋白质峰在痰瘀互结组中高表达,BPS判别分析选出M/Z为4099.85、9418.17的2个蛋白质峰构建两组间血清差异蛋白决策树分类诊断模型.结论:初步建立了甲状腺癌的中医辨证模式——血清蛋白质指纹图谱,也为中医证候实质的研究探讨新的方法和途径.Objective:To investigate the difference of serum protein mass spectrum between different TCM syndromes of thyroid cancer,and establish a diagnostic model.Methods:Serum protein fingerprints of 40 thyroid cancer patients (19 patients of Liver qi stagnation syndrome,21 patients of phlegm and blood stasis syndrome)were detected by using SELDI-TOF-MS (Surface enhanced laser desorption/ionization time-of-flight mass spectrometry),differentially expressed proteins between the two groups were identified,and a diagnostic model was established with the help of bioinformatics.Results:86 protein peaks were detected within the range from 2000Da to 20000Da,22 of which showed significant difference between the two groups (P < 0.01).17 of the 22protein peaks were highly expressed in the group of liver qi stagnation,and 5 were highly expressed in the group of phlegm and blood stasis.Two protein peaks (M/Z =4099.85,9418.17) were selected by Biomarker Pattern Software (BPS) to construct a decision-tree classification diagnostic model of differentially expressed proteins in serum.Conclusion:A preliminary TCM syndrome differentiation model-serum protein fingerprint was established,which provides a novel method and approach to the investigation of the essence of TCM syndromes.
