摘 要:目的探讨眼镜蛇毒C组分对白血病细胞基质金属蛋白酶(MMP)-2和MMP-9表达和活性的影响。方法用10 ng/mL的眼镜蛇毒C组分对白血病细胞进行0、12、24、48 h处理。用ELISA、明胶酶谱法和荧光RT-PCR法检测不同时间处理后白血病细胞MMP-2和MMP-9的表达水平。结果与处理0 h相比,白血病细胞经10 ng/mL的眼镜蛇毒C组分分别处理12、24、48 h后,用ELISA检测结果显示MMP-2、MMP-9活性下降,差异有统计学意义(P<0.05,P<0.01);明胶酶谱法检测结果显示MMP-2、MMP-9活性下降,差异有统计学意义(P<0.05,P<0.01);荧光定量RT-PCR检测结果显示,MMP-2、MMP-9 mRNA相对含量下降,差异有统计学意义(P<0.05,P<0.01)。结论 10 ng/mL眼镜蛇毒C组分对白血病细胞MMP-2和MMP-9的表达均有抑制作用,眼镜蛇毒C组分对白血病细胞的抑制作用可能与其抑制MMP-2和MMP-9表达和活性相关。Objective To investigate the effects of snake venom protein fraction C of Naja atra on the activities and expression of matrix metalloproteinase (MMP) - 2 and MMP - 9 in leukemia cells. Methods MMP - 2 and MMP - 9 activities in leukemia cells were assessed by gelatin zymography and ELISA after treated with 10 ng/mL of snake venom protein fraction C for different duration (0, 12, 24 and 48 h). The expressions of MMP-2 and MMP-9 mRNA in leu- kemia cells were assessed by fluorescent quantitative RT -PCR. Results Compared with control group (baseline), the activities of MMP -2 and MMP -9 in leukemia cells treated for 12, 24 and 48 h were significantly reduced according to both ELISA and gelatin zymography test ( P 〈 0. 05 ). Meanwhile, significant down - regulation of MMP - 2 and MMP - 9 mRNA (P 〈 0. 05) in leukemia cells treated with snake venom protein fraction C was observed according to fluorescent quantitative RT - PCR. Conclusion The expressions and activities of MMP - 2 and MMP - 9 are reduced by snake ven-om protein fraction C, which may be responsible for the inhibitory effects on leukemia cells.
