丙戊酸钠联合三氧化二砷对NB4细胞株血管内皮生长因子及其受体表达的影响

摘  要:目的探讨丙戊酸钠(VPA)联合三氧化二砷(As2O3)对急性早幼粒细胞白血病(acute promyelocytic leukemia,APL)NB4细胞株血管内皮生长因子(VEGF)及其受体Flt-1表达的影响。方法实验分空白对照组、As2O3组、As2O3+VPA组,采用反转录聚合酶链反应方法检测VEGF、Flt-1 mRNA的表达,应用蛋白印迹法(Western-blot)检测VEGF、Flt-1蛋白的表达。结果 NB4细胞在As2O3作用下VEGF mRNA、VEGF蛋白、Flt-1mRNA和Flt-1蛋白表达水平明显增高,分别为VEGF mRNA 0.426±0.026vs 0.268±0.023,VEGF蛋白0.648±0.027vs 0.588±0.031,Flt-1mRNA 0.922±0.032vs 0.766±0.030,Flt-1蛋白0.520±0.032vs 0.320±0.036(P<0.05或<0.01)。As2O3+VPA组VEGF mRNA、VEGF蛋白、Flt-1mRNA和Flt-1蛋白比对照组和As2O3组均明显降低(均P<0.01)。VEGF mRNA表达水平降至0.129±0.028,VEGF蛋白表达水平降至0.328±0.029,Flt-1mRNA表达水平降至0.522±0.025,Flt-1蛋白表达水平降至0.124±0.024。结论 As2O3诱导NB4细胞VEGF、Flt-1表达增高,VPA可下调As2O3引起的VEGF、Flt-1表达增高。Objective To investigate the expressions of vascular endothelial growth factor, Fit-1 in NB4 cells lines affected by valproate sodium (VPA) combining with arsenic trioxide (As2 Oa ). Methods NB4 cells lines were treated with As2 O3 ,As2 Oa in combination with VPA and control group. The effect on mRNA expression of VEGF and Flt-1 was detected by reverse transcription-polymerasechain reaction. The protein expression of VEGF and Flt-1 were measured by Western blot. Results Compared with control group, the mRNA and protein expression of VEGF, Fit-1 increased signi{icantly under Ass Oa e{{ect, the mRNA expression of VEGF 0. 426 ± 0. 026 vs 0. 268 ± 0. 023, the protein expression of VEGF 0. 648±0. 027 vs 0. 588±0. 031,the mRNA expression of Flt-1 0. 922±0. 032 vs 0. 766±0. 030, the protein expression of Flt-1 0. 520±0. 032 vs 0. 320±0. 036( P 〈0.05 or 〈0.01). Compared with control group and As2O3 group, the mRNA and protein expression of VEGF,Flt-1 in NB4 cells of As2 O3 in combination with VPA group were all obviously decreased( P 〈 0.01). The mRNA and protein expression of VEGF decreased to 0. 129 ± 0. 028,0. 328±0. 029,the mRNA and protein expression of Fh 1 decreased to 0. 522±0. 025,0. 124±0. 024. Conclusion As2O3 can induce the expression of VEGF, Flt-1. VPA can down-regulate the increased expression of VEGF, Fh 1caused by As2O3.
