摘 要:近年来,动物细胞培养技术在生物制药领域成为最受关注的热点之一,灌注培养技术广泛应用于动物细胞高密度培养,推动了现代生物医药产业的发展。通过对动物细胞代谢分析引出灌注培养优势特点,再介绍常规灌注培养系统组成结构,对其核心装置细胞培养罐和截留装置进行综述。其中重点分析了适用于动物细胞悬浮培养灌注系统的流程原理及灌注速率控制方法,Elephant Ear桨的性能研究以及旋转过滤器的结构设计。总结近年来国内外文献关于灌注培养在组织工程、疫苗和单克隆抗体方面的应用。最后指出研究灌注系统存在的难点,展望了灌注培养系统的应用前景。In recent years,animal cell culture technology becomes one of the most popular hot spots in the field of bio-pharmaceutics, perfusion culture techniques are widely used in animal cell density culture, promote the development of the modern biological medicine industry. From the analysis of animal cell metabolism,the perfusion culture advantages and the structure of conventional perfusion culture system were introduced,its core unit cell culture vessel and intercept device were reviewed. The process of perfusion system which is suitable for animal cell suspension culture and perfusion rate control method, Elephant Ear oar research as well as the structure of rotating filter designing were analyzed and emphasized. The recent literature at home and abroad about perfusion culture in tissue engineering, vaccine and monoclonal antibody were summarized. Finally, the difficulties existing in the system of perfusion,and the application prospect of perfusion culture system were pointed out.
