乳腺特异的甜味蛋白Brazzein基因真核表达载体的构建

摘　　要：为了建立通过山羊乳汁获取甜味蛋白Brazzein的方法,研究在人工合成甜味蛋白Brazzein基因的基础上,利用乳腺特异表达的pBC-1载体,构建Brazzein基因的真核表达载体。结果表明:所合成的Brazzein基因与GenBank中Brazzein序列的同源性为100%,构建的克隆载体STP-pMD18-Simple正确;将甜味蛋白Brazzein基因与pBC-1载体连接后,成功构建甜味蛋白Brazzein基因的乳腺特异性表达载体STP-pBC-1。To establish the method of obtaining sweet protein Braszein through goat milk, on the basis of synthetic sweet protein Brazzein gone, an eukaryotic expression Vector for sweet protein Brazzein gone was constructed using mammary gland - specific expression vector pBC - 1. The results showed that the sequeneeof the synthesized Brazzein gene shared 100 percent homology with that of published in GenBank, and the con- structed cloning vector STP -pMD18 -Simple was correct. After the sweet protein Brazzein gone was connected with pBC -1 vector, the mam- mary gland - specific expression vector STP - pBC - 1 for sweet protein Brazzein gone was successfully constructed.
