摘 要:目的观察大鼠脊髓损伤模型中磷酸化信号转导与转录激活因子3(p-STAT3)及Th17相关因子的水平,初步探讨STAT3对脊髓损伤后Th17细胞的作用。方法成年健康雄性SD大鼠75只随机分为对照组(n=15)和模型组(每个时点n=15)。模型组采用改良Allen氏打击法建立脊髓损伤模型,对照组仅行椎板全切除术。在建模后1、24、48和72 h获取模型组大鼠损伤段脊髓、脾脏及外周血,免疫组织化学染色法检测白细胞介素(IL)-17在损伤段脊髓组织中的分布及表达情况,蛋白免疫印迹法检测p-STAT3在损伤段脊髓和脾脏组织中的表达变化,ELISA法检测外周血IL-6、IL-21和IL-23的动态变化,并与对照组大鼠作比较。结果模型组各时点IL-17、p-STAT3、IL-6、IL-21和IL-23的表达明显高于对照组(P<0.05),相关性分析显示p-STAT3的表达均与IL-17、IL-6及IL-21的表达呈正相关性(P<0.05)。结论 STAT3的异常激活可能是造成脊髓损伤后Th17细胞过度表达的重要原因。Objective To investigate the expression of p -STAT3(signal transducer and activator of transcription 3) and Th17 cell related signal molecules in the rat model of spinal cord injury , so as to explore the roles of STAT3 and its related signal molecules in spinal cord injury .Methods A total of 75 adult male SD rats were randomly divided into two groups, control group (n=15) and model group (n=15 each time point).Acute spinal cord injury rat model was estab-lished by the modified Allen weight drop method in the model group , while laminectomy alone was performed in control group.Each 15 rats in model group were sacrificed at the 1st, 24th, 48th and 72nd hour after operation for collection of spi-nal cord tissues, spleen tissues, and peripheral blood.Immunohistochemistry method was used to detect the expressions of interleukin ( IL)-17 in damaged spinal cord tissues , Western blot was applied to assess the protein expression of p -STAT3 in the spinal cord and spleen tissues , and ELISA was used to measure serum IL -6, IL-21 and IL-23.Results Expression of IL-17, p-STAT3, IL-6, IL-21 and IL-23 in model group was significantly higher than control group (P〈0.05).Furthermore, the expression of p -STAT3 was significantly positively correlated with IL -17, IL-6 and IL-21 (P〈0.05).Conclusion Aberrant activation of STAT3 may be one of factors to contribute to Th17 polariza-tion after spinal cord injury .
