两种中药复方对局灶性脑缺血再灌注大鼠额顶叶皮质LRP5和β-Catenin表达的影响

摘　　要：目的:观察补肾生髓方和益气活血方对局灶性脑缺血再灌注大鼠额顶叶皮质LRP5和β-Catenin蛋白表达的影响。方法:大鼠随机分为假手术组、模型组、补肾生髓方组、益气活血方组。线栓法阻塞大鼠左侧大脑中动脉,制作局灶性脑缺血再灌注大鼠模型,脑缺血2?h再灌注2周。治疗组连续给药2周,采用免疫组化EnVision两步法检测额顶叶皮质LRP5和β-Catenin表达的阳性面积和平均吸收光密度值。结果:皮质LRP5的平均吸收光密度值(A值),模型组与假手术组之间无明显差异,模型组与补肾组差异显著(P<0.05),模型组与益气组差异不明显,补肾组与益气组之间有明显差异(P<0.01);皮质LRP5的阳性面积值(Area值),模型组与假手术组差异不显著,与各治疗组之间有明显差异(P<0.05),补肾组与益气组之间无明显差异。皮质β-Catenin的A值,模型组与假手术组之间有明显差异(P<0.05),益气组、补肾组与模型组之间无明显差异,补肾组与益气组之间无明显差异;皮质β-Catenin的Area值,模型组与假手术组之间无明显差异,与各治疗组之间差异具有显著性(P<0.05),补肾组与益气组之间无明显差异。结论:益气活血方和补肾生髓方能通过降低额顶叶皮质LRP5和升高β-Catenin蛋白表达,调节Wnt信号转导通路,促进脑缺血再灌注后内源性神经干细胞的增殖分化。Objective ： To investigate the influences of Yiqi Huoxue Formula （ YF ） and Bushen Shengsui Formula （ BF ） on the expressions of LRP5 and β-Catenin cortical in cerebral tissue after focal ischemia-reperfusion （ I/R ） in rats, and to investigate the mechanism of those resisting on loealcerebral ischemia and reperfusion inflammatory injury. Methods ： The animals were divided into the sham-operative group, the model group, the YF group and the BF group. The local cerebral I/R model was established by the ligation of the middle cerebral arteries by using thead approach. After cerebral ischemia for 2 hours, the reperfusion was given to the rats for 2 weeks in treatment groups respectively. The positive area and the value of average absorbance （A value ） in the expressions of LRP5 and β-Catenin were detected by using immunofluorescence staining method in ischemic penumbra. Resuhs ： The average absorption optical density of cortical LRP5 values （ A values ） had no obvious difference between the model group and the sham- operative group and there was a significant difference between the BF group and model group （P〈0.05）. There was no significant difference between the treatment groups and the model group. There was an obvious difference between the BF group and the YF group （P〈0.01）. The positive area of cortical LRP5 value （ Area value ） had no obvious difference between the model group and the sham-operative group and there was a significant difference between the treatment group and model group （P〈0.05）and no significant difference between the YF group and the BF g：oup, For the Cortical β-Catenin A value, there was an obvious difference between the model group and the sham-operative group （ P〈0.05 ） and there was no significant difference between the two treatment groups and model group and no obvious difference between the model group and the sham-operative group.
