ezrin蛋白在青海撒拉族和汉族胃癌中的表达及其生物学功能研究

摘　　要：目的对ezrin蛋白在青海撒拉族和汉族胃癌中的表达及其生物学功能行相关研究。方法用免疫组化法检测ezrin蛋白在胃癌组织和正常胃粘膜组织中的表达;MTT法检测胃癌细胞增殖;双荧光素酶及Western blot蛋白印迹法检测酶活性和蛋白表达情况。结果 ezrin蛋白在青海撒拉族和汉族胃癌组织中表达比正常胃黏膜组织高(P<0.05),且均与胃癌分化程度、淋巴结转移有关(P<0.05),而和年龄、性别无关(P>0.05)。ezrin蛋白对胃癌细胞增殖具有一定抑制能力,同时可以增加由RhoA(Ac)激活型诱导的荧光酶素。结论 ezrin蛋白表达水平与青海撒拉族和汉族胃癌易感性相关,且参与调节由RhoA诱导的细胞内信号途径,从而调节细胞极化、细胞骨架重组等功能。Objective To investigate ezrin protein expression in gastric cancer and its biological functions in Qinghai Salar and Han nationalities.Method Ezrin protein expression in gastric cancer and normal gastric mucosa was detected by immunochemical staining.MTT was used to analyze the cell proliferation.Dual-fluorescent assay and Western blot were used to detect the protein activity and expression.Results Ezrin protein expression was higher in gastric cancer than normal gastric mucosa in both Qinghai Salar and Han patients（P〈0.05）.The expression level of ezrin protein was correlated with the degree of tumor differentiation and lymph node metastasis of Qinghai Salar and Han patients（P〈0.05）,and it is not related with ages and gender of patients in both Salar and Han nationalities（P〉0.05）.Ezrin showed an inhibitive effect on the proliferation of gastric cancer cells,and it can enhance the fluorescent activity induced by RhoA（Ac）,which is constitutively active.Conclusion The expression of ezrin is related with the susceptibility to gastric cancer development and metastasis in both Qinghai Salar and Han patients,and it is involved in regulation of cell polarization,cytoskeleton remodeling through RhoA-mediated signal pathway.
