摘 要:目的:探讨香芹酚(carvacrol,CV)对雄激素非依赖型前列腺癌DU145细胞株的抗增殖、诱导凋亡作用及相关分子机制。方法:采用不同浓度的CV(0、10、20、40、80、100μmol/L)体外处理DU145细胞24 h或48 h;采用四甲基偶氮唑蓝(methyl thiazolyl tetrazolium,MTT)法检测不同浓度CV对DU145细胞增殖的抑制率;采用Hoechst染色法及流式细胞术检测细胞凋亡情况;采用逆转录聚合酶链反应(reverse transcription-polymerase chain reaction,RT-PCR)及蛋白质印迹法(Western blotting)检测凋亡相关基因及蛋白表达水平。结果:CV对前列腺癌DU145细胞的增殖有明显的抑制作用,且呈剂量及时间依赖性;Hoechst染色法及Annexin V-FITC/PI双染法检测结果显示,不同浓度香芹酚处理DU145后,细胞凋亡程度及比率均明显增加;RT-PCR及Western blotting检测结果显示,随着CV浓度的增加,DU145细胞中Bcl-2表达水平降低,而Bax及Caspase3表达水平升高,细胞色素C(cytochrome C,Cyt C)在细胞质中的释放增加(P<0.05)。结论:CV能够抑制DU145细胞的增殖并诱导其凋亡,其机制与调控Bcl2家族蛋白和Caspase3的表达以及Cyt C的释放等有关,CV在雄激素非依赖型前列腺癌的治疗中具有潜在的应用价值。Objective: To investigate the anti-proliferative and apoptosis-inducing effects of carvacrol (CV) in androgen inde- pendent prostate cancer cell line DU145 and its molecular mechanisms. Methods:DU145 cells were incubated in various concen- trations of CV (0,10,20,40,80,100 μmol/L) in vitro for 24 h or 48 h. Inhibition rates of different concentrations of CV on proliferation of DU145 cells were detected by methyl thiazolyl tetrazolium(MTT) assay. Cell apoptosis was detected by Hoeehst staining and flow cytometry. Reverse transcription-polymerase chain reaction (RT-PCR) and Western blotting were used for de- tection of apoptosis related genes and protein expression levels. Results: CV had a significant anti-tumor effect on prostate cancer cell line DU145 in a dose-dependent and time-dependent manner. Hoeehst and Annexin V FITC/PI results showed that the degrees and ratios of apoptosis were significantly increased after treating by different concentrations of CV. RT-PCR and Western blotting results showed that Bcl-2 expression was decreased, Bax and Caspase3 expression were increased, while the release of cytochrome C(Cyt C) in the cytoplasm of DU/45 cells was increased with the increase of the concentration of CV (P 〈0.05). Conclusions: CV can inhibit DU145 cell proliferation and induce apoptosis by regulating the expression of BeI2 family and Caspase3 and regulating Cyt C releasing. CV has potential application value in the treatment of androgen-independent prostate cancer.
